摘  要:牛干灰皮胶原纤维先经回湿、酸溶胀和磨浆处理,再分别与不同浓度(0.2 wt%、1.0 wt%和1.8 wt%)的茶多酚和30 wt%的甘油充分分散(均基于干胶原质量),采用溶液铸膜法制备了系列茶多酚改性胶原复合膜(CCF).用傅里叶变换红外光谱仪、X射线衍射仪、差失扫描量热仪、水接触角测量仪、扫描电镜及拉伸试验机等仪器对CCF的结构、热性能、亲疏水性、微观形貌和机械性能进行研究.结果表明,CCF内保持了三股螺旋胶原结构的完整性但又以无定形态为主.随着茶多酚用量的增加,CCF内胶原肽链主要以无定形分布且肽链间横向堆积距离有变大趋势,CCF的热稳定性增加.扫描电镜测试结果表明,不加茶多酚的对照膜表面有空洞,不平整,膜断面不紧实;随着茶多酚用量的增加,CCF表面的空洞消失且颗粒数量逐渐增加,膜断面的紧实度也不断增大.CCF表面的静态水接触角由对照组的93.4°逐渐增加至124.3°,表明疏水性增加.膜抗张强度也由5.17 MPa增加至9.78 MPa,延伸率由6.1%降至2.5%.因此,可为牛干灰皮胶原纤维的资源化利用带来新的契机.Dry limed cowhide collagen fibers were rewetted,acid-swollen and ground to obtain a homogeneous collagen syrup.Different concentrations of tea polyphenols(0.2 wt%,1.0 wt%and 1.8 wt%)and glycerol(30 wt%,all based on dry collagen fibers)were orderly added into the above collagen syrup and fully dispersed to form the film-forming solutions.Finally a series of collagen composite films(CCF)were prepared by solution casting method.The structure,thermal properties,hydrophilicity,microstructure and mechanical properties of CCF were studied by Fourier transform infrared spectrometer(FT-IR),X-ray diffrotometer(XRD),differential scanning calorimetry(DSC),water contact angle meter,scanning electron microscope(SEM)and tensile testing machine.Results showed that the integrity of the three-strand spiral collagen structure was maintained in CCF,but the amorphous structure was dominant.With the increase of the amount of tea polyphenols(TP),the collagen molecular chains in CCF film were mainly distributed in an amorphous form,and the intermolecular lateral packing distance among the peptide chains of triple helix tended to increase,and thermal stability of CCF film increased.Scanning electron microscopy(SEM)results showed that the control film surface without TP was not smooth with some cavities,and the film cross-section was not compact.With the increase of the TP amount,the voids on the CCF surface disappeared and the number of particles gradually increased,and the compactness of the film section also increased.The static water contact angle of the film surface increased from 93.4°to 124.3°,indicating the increased hydrophobicity.The tensile strength of the film increased from 5.17 Mpa to 9.78 Mpa,and the elongation decreased from 6.1%to 2.5%.Therefore,this study can bring a new opportunity for the resource utilization of dry limed cowhide collagen fibers.
