摘  要:对山茱萸粗多糖超声辅助法提取工艺进行优化并研究其在体外对α-葡萄糖苷酶的抑制作用,考察其降血糖活性的潜能。以粗多糖提取率为考察指标,通过单因素试验及正交试验对山茱萸粗多糖的提取条件进行优化并验证。通过PNPG法以阿卡波糖为阳性对照测定山茱萸粗多糖对α-葡萄糖苷酶的抑制作用。结果表明,山茱萸粗多糖提取最佳条件为超声温度60℃、超声功率450 W、超声时间50 min、料液比1︰30 g/mL。在此条件下,山茱萸粗多糖的提取率为30.43%。10 mg/mL的山茱萸粗多糖对α-葡萄糖苷酶活性的抑制率达70%,具有降血糖活性的潜能。Aim to optimize the ultrasonic-assisted extraction method of crude polysaccharide from Cornus officinalis.Its inhibition effect on α-glucosidase in vitro as well as its potential of hypoglycemic activity was studied.Single factor test and orthogonal test were performed to optimize the extraction condition of the crude polysaccharide using extraction efficiency as index.Then the inhibition ofα-glucosidase by extracted crude polysaccharide was determined by PNPG method with acarbose as the positive control.The results showed that the optimum condition was temperature 60℃,power 450 W,time 50 min,and material-liquid ratio of 1︰30 g/mL.The extraction efficiency of crude polysaccharide was 30.43% under this condition.The inhibition ratio of α-glucosidase by 10 mg/mL crude polysaccharide was 70%.It is suspected that crude polysaccharide from Cornus officinalis might have a potential hypoglycemic effect.
