摘 要:以甘薯生全粉为原料,采用米曲酶发酵提取去氢表雄酮(dehydroepiandrosterone,DHEA),基于提取物中DHEA定性分析基础上,探讨米曲酶孢子悬浮液接种量、发酵时间、发酵温度和液固比等因素对米曲霉发酵提取甘薯中DHEA得率的影响,结合单因素与响应面试验优化得最优发酵工艺:米曲酶孢子悬浮液接种量6%,发酵时间72 h,发酵温度30℃,液固比15∶1(mL/g),此条件下DHEA得率可达94.70μg/100 g,显著(p<0.05)高于预发酵-酸解法、酶解法提取DHEA得率。Using whole sweet potato powder as the raw material and on the basis of pre-testing the identification of the sweet potato fermentation extract,four factors(fermentation liquid ratio,fermentation time,inoculum amount,and fermentation temperature)were studied in the extraction of dehydroepiandrosterone(DHEA)from sweet potato through fermentation using Aspergillus oryzae.Combined with response surface analysis,the optimal technological conditions for extracting DHEA from sweet potato by Aspergillus oryzae fermentation were revealed as:fermentation material to liquid ratio of 1∶15(g/mL),fermentation time 72 h,inoculum 6%,and fermentation temperature 30℃.Under these conditions,the DHEA yield reached 94.70μg/100 g,which was close to the theoretical prediction value of 97.07μg/100 g.Compared with the existing pre-fermentation-acid hydrolysis method,enzymatic hydrolysis significantly increased DHEA yield(p<0.05).
