摘 要:本文建立了一种采用高效液相色谱法同时测定鸡精调味料中的黄色合成着色剂(柠檬黄、日落黄)含量的方法。样品经提取后通过高效液相色谱DAD检测器分析,以色谱峰保留时间和DAD光谱进行定性,采用C_(18)色谱柱,以甲醇-乙酸铵(0.02 mol/L)为流动相,梯度洗脱,进样量10μL,检测波长428 nm(0~6 min测定柠檬黄)、484 nm(6~25 min测定日落黄),峰面积外标法定量。结果表明,2种着色剂在1~50μg/mL有良好的线性关系,相关系数R^(2)均大于0.999,方法检出限为柠檬黄0.15 mg/kg、日落黄0.15 mg/kg,加标回收率可达93.0%~100.1%,相对标准偏差均不大于2.3%。该方法简便、准确度高、重复性好,为进一步控制鸡精的质量标准提供了依据。In this test,a method for simultaneous determination of yellow synthetic colorant(tartrazine,sunset yellow)in chichen essence seasoning by high performance liquid chromatography was established.After extraction,the samples were analyzed by high performance liquid chromatography dad detector.The chromatographic peak retention time and DAD spectrum were used for qualitative analysis.C^(18) chromatographic column was used,methanol ammonium acetate(0.02 mol/L)was used as mobile phase,gradient elution,and the injection amount was 10μL.The detection wavelength was 428 nm(lemon yellow was measured in 0~6 min)and 484 nm(sunset yellow was measured in 6~25 min).The peak area was quantified by external standard method.The results showed that there was a good linear relationship for the two colorants in the range of 1~50μg/mL,and the correlation coefficients(R2)were greater than 0.999.The detection limit of tartrazine was 0.15 mg/kg and sunset yellow was 0.15 mg/kg.The recovery rate of standard addition was 93.0%~100.1%,and the relative standard deviation was less than 2.3%.The method is simple,accurate and repeatable,and provides a basis for further control of the quality standard of chicken essence.
