摘 要:通过跟踪分析不同生产排次、窖内不同位置和不同发酵时间节点的酒醅中正丁醇的生成规律,明确了窖泥是浓香型白酒中产正丁醇微生物的重要载体;通过对筛自窖泥的8株梭菌进行产正丁醇能力评价,表明C.beijerinckii产丁醇能力最强,继而以荧光定量PCR法进一步证实C.beijerinckii是浓香型白酒发酵过程中正丁醇合成的主要微生物;通过设计添加糖、有机酸和氨基酸等三类9种物质,证实了C.beijerinckii是以丁酸和葡萄糖为主要前体物质,通过糖代谢合成途径(Harris途径)生成正丁醇,这为调控浓香型白酒正丁醇含量提供了理论依据。Through tracking the generation of n-butanol in the fermented grains of different production cycles,at different positions in the pit and at different fermentation stages,it is found that the pit mud is an important carrier of n-butanol-producing microorganisms.The n-butanol-producing capacity of 8 Clostridium strains screened from the pit mud is evaluated,and C.beijerinckii is found to have the highest n-butanol-producing capacity.Then qRT-PCR analysis is carried out,and C.beijerinckii is proved to be the main n-butanol-producing microorganism in the fermentation process of Nongxiang Baijiu.Precursor(carbon sources,organic acids and amino acids)addition tests are carried out,and it is proved that C.beijerinckii uses butyric acid and glucose as the main precursors to produce n-butanol through the Harris pathway.This study has provided a theoretical basis for the regulation of n-butanol content in Nongxiang Baijiu.
