摘 要:以树莓果渣多酚为还原剂,硝酸银为银源,采用超声波辅助生物还原法制备多酚纳米银。首先利用紫外-可见分光光度计和Nano ZS90粒度分析仪,通过单因素实验探讨了不同硝酸银与多酚质量比、溶液pH值以及超声时间对合成纳米银紫外-可见吸收光谱和粒径的影响。确定了超声波法制备多酚纳米银的最佳条件:硝酸银与多酚质量比为1:2,溶液pH值为10.5,超声时间10 min,此时多酚纳米银平均粒径为12.55 nm。之后,采用傅立叶变换红外光谱仪、透射电子显微镜、X射线衍射仪、X射线光电子能谱仪对多酚纳米银结构进行表征,结果显示:树莓果渣多酚可还原硝酸银,得到分散均匀、近圆球状的面心立方结构纳米银颗粒,银原子含量为1.52%。对3种细菌(金黄色葡萄球菌、大肠杆菌、沙门氏菌)的抑菌活性初步测定结果表明,多酚纳米银能有效地抑制3种细菌的生长,其中对金黄色葡萄球菌的抑制作用最明显,最小抑菌浓度和最小杀菌浓度分别为0.156 mg/mL和0.313 mg/mL。该研究为树莓果渣多酚的进一步利用奠定了理论基础。Silver nanoparticles(AgNPs)were prepared by ultrasonic assisted biological reduction method with raspberry pomace polyphenol as reducing agent and silver nitrate as silver source.Through single factor experiment,the effects of the factors,including the mass ratio of silver nitrate to polyphenols,pH,ultrasonic time on the ultraviolet-visible absorption spectrum and particle size of AgNPs were evaluated by ultraviolet-visible spectrophotometry and Nano ZS90 particle size and potential analyzer.The optimal reaction conditions were obtained as follows:mass ratio of AgNOto polyphenols 1:2,pH 10.5,ultrasonic time 10 min,the average particle size of polyphenol AgNPs was 12.55 nm.The structure of polyphenol AgNPs was characterized by fourier transform infrared spectroscopy,transmission electron microscopy,X-ray diffractometer,X-ray photoelectron spectroscopy.The results showed that AgNOcould be reduced by the raspberry pomace polyphenol,and the polyphenol AgNPs presented uniform dispersion,face-centered cubic and nearly spherical shapes,and the silver content reached 1.52%.The results of antibacterial activities against the three bacterias(Staphylococcus aureus,Escherichia coli,Salmonella)demonstrated that polyphenol AgNPs could significantly inhibit the growth of the three bacterias.Among them,Staphylococcus aureus was most susceptible to polyphenol AgNPs,and the minimum inhibitory concentration and minimum bactericidal concentration were 0.156 mg/mL and 0.313 mg/mL,respectively.This study provided a scientific theoretical basis for the further development and utilization of raspberry pomace.
