摘  要:为高效利用绿碎茶资源,采用沸水浸提法与冻干法制备绿碎茶水提物,测定其总酚、总黄酮、总糖以及酸性多糖含量,通过DPPH、ABTS、HRSA、FIC和FRAP五种方法评价其体外抗氧化活性,并研究其对α-葡萄糖苷酶与乙酰胆碱酯酶的抑制活性。结果表明,绿碎茶水提物总糖与酸性多糖、总黄酮及总酚含量分别为256.27mg GE/g、105.45mg AGE/g、218.45mg RE/g及203.72mg GAE/g。绿碎茶水提物对DPPH·、ABTS^(+)·、·OH清除率以及FIC和FRAP的IC_(50)值分别为76.20、22.16、2952.57、643.81及86.86mg/L。绿碎茶水提物对α-葡萄糖苷酶抑制能力(IC_(50)值为0.39mg/mL)强于阿卡波糖(IC_(50)值为0.60mg/mL)。绿碎茶水提物对乙酰胆碱酯酶也具有一定的抑制能力,IC_(50)值为2.70mg/mL。因此,绿碎茶水提物具有植物源天然抗氧化剂及α-葡萄糖苷酶与乙酰胆碱酯酶抑制剂的开发潜力。The total polyphenol,toal flavonoid,total sugar and acidic polysaccharide in the broken green tea extract(BGTE)obtained by boiling water extraction and lyophilization were measured,and the in vitro antioxidant activities were evaluated by five methods including DPPH,ABTS,HRSA,FIC and FRAP assays,and the inhibitory activities againstα-glucosidase and acetylcholinesterase were also investigated for efficient utilization of the broken green tea resource in this study.The results showed that the contents of the total polyphenol,toal flavonoid,total sugar and acidic polysaccharide in BGTE were 203.72mg GAE/g,218.45mg RE/g,256.27mg GE/g and 105.45mg AGE/g,respectively.BGTE displayed good antioxidant activity with the IC_(50) values of DPPH,ABTS,HRSA,FIC and FRAP of 76.20,22.16,2952.57,643.81 and 86.86mg/L,respectively.Furthermore,BGTE exhibited stronger inhibitory ability againstα-glucosidase with the IC_(50) value of 0.39mg/mL compared to acarbose(0.60mg/mL).Moreover,BGTE showed certain inhibitory activity against acetylcholinesterase with the IC_(50) value of 2.70mg/mL.Therefore,the results suggest that the broken green tea extract is a good source of potential natural antioxidants andα-glucosidase and acetylcholinesterase inhibitors.
