摘　　要：研究旨在从健康犬粪便中分离筛选具有缓解犬腹泻功能的乳酸菌。通过革兰氏染色、16S rDNA序列分析对健康犬粪便中分离的30株菌进行鉴定,对菌株抑菌能力、抗生素敏感性、耐酸耐胆盐能力及细胞黏附能力进行评价,选出最佳菌株进行动物试验。结果显示:J40、J48等9株乳酸菌对大肠杆菌、志贺氏菌、沙门氏菌和空肠弯曲杆菌等具有较好抑菌效果,J40抑菌效果最强。抗生素敏感性试验显示:9株乳酸菌对青霉素、氨苄西林及头孢噻吩等3种抗生素均高度敏感,表明9株菌株不携带上述耐药基因,具有一定安全性和可控性。试验筛选的4株菌株在pH值2、胆盐浓度0.1%条件下培养2 h的存活率均不低于50%,具有较好的耐酸耐胆盐能力;4株菌对Caco-2细胞的黏附个数均大于6,J40黏附性能最佳。选用J40作为治疗组对志贺氏菌攻毒幼犬进行体内试验,结果显示:菌株J40可有效降低幼犬腹泻率,治疗细菌性腹泻。研究表明,J40可作为宠物用微生态制剂的潜在益生菌株。The study was to isolate and screen lactic acid bacteria with potential probiotics from feces of healthy canine.In the study,30 strains isolated from the feces of healthy canine were identified by Gram staining,colony morphology observation and 16S rDNA sequence analysis,and the functional evaluation of bacteriostatic ability,antibiotic sensitivity,acid tolerance and bile salt tolerance,cell adhesion ability were carried out.The best strains were selected for animal experiment.The results showed that nine strains of Lactobacillus had good bacteriostatic effect on Escherichia coli,Shigella,Salmonella and Campylobacter jejuni,among which strain J40 had the strongest bacteriostatic effect.Antibiotic sensitivity tests showed that nine lactic acid bacteria strains were highly sensitive to penicillin,ampicillin and cephalothiophene antibiotics,which proved that the nine strains did not carry the drug resistance genes,which had certain safety and controllable.The survival rate of four strains cultured at pH value 2 and bile salt concentration 0.1%for 2 h were all no less than 50%,showing good acid tolerance and bile salt tolerance.The number of adhesion to Caco-2 cells by four strains above was all more than six,and J40 had the best adhesion performance.J40 was selected as the treatment group to conduct in vivo experiments on Shigella challenged puppies.The results showed that strain J40 could effectively reduce the rate of diarrhea in puppies,treat bacterial diarrhea.The experiment indicates that J40 can be used as a potential probiotic strain for pet microecologics.
