摘  要:为了扩充嗜热芽孢杆菌基因组数据库,进一步筛选可用于微生物抑制法检测药物残留的候选菌株,本试验对2株嗜热芽孢杆菌(Commercial isolate和CAU475)进行全基因组和耐药性分析。结果显示,经16S rRNA基因序列分析及进化树构建,判定2株分离菌株为嗜热脂肪芽孢杆菌;经核心基因组进化树及全基因组相似性比对分析,2株分离菌株与用于抗生素残留检测指示菌株的Geobacillus stearothermophilus ATCC 7953同源性较高,推测具有相似的生物学特性;通过对Commercial isolate中携带的1个大小为44.8 kb质粒的49个开放阅读框进行标注与分析,丰富了嗜热脂肪芽孢杆菌的质粒基因信息;药敏试验结果显示,2株分离菌株对10种常用抗菌药物均敏感。综上所述,2株嗜热脂肪芽孢杆菌可作为候选菌株用于微生物抑制法检测药物残留。In order to expand the genome database of Bacillus thermophilus and further screen the candidates that can be used for the detection of drug residues by microbial inhibition,the whole genomes and drug resistance of two Bacillus thermophilus strains(Commercial isolate and CAU475)were analyzed.The results showed that these two strains were identified as Geobacillus stearothermophilus by 16 S rRNA sequencing and phylogenetic tree analysis,and further confirmed by their high genome homology with Geobacillus stearothermophilus ATCC 7953.Sequencing analysis of 49 ORFs in the 44.8 kb plasmid of Geobacillus stearothermophilus commercial strain revealed additional genetic details.The two Geobacillus stearothermophilus strains were sensitive to 10 antibiotics tested.Thus,these two Geobacillus stearothermophilus strains can be used to detect drug residues by microbial inhibition.
