摘 要:以珊瑚菌液体培养菌丝体为原料,基于单因素试验,利用响应面法优化珊瑚菌菌丝体多糖提取工艺,同时对提取的菌丝体多糖结构进行初步分析。结果表明:优化后的最佳提取工艺条件为料液比1∶25(g/mL)、提取温度83℃、提取时间4.2 h。在此条件下,菌丝体多糖得率平均为(15.10±0.26)%。高效液相色谱法(HPLC)柱前衍生化分析菌丝体多糖的单糖组成,表明其包含甘露糖、核糖、鼠李糖、葡萄糖、半乳糖、木糖和阿拉伯糖,对应摩尔比为0.12∶0.55∶0.41∶1.00∶0.20∶1.15∶0.82;高效凝胶渗透色谱法分析表明其主要包括2种多糖组分,分子质量分别为9.65×10^(5)、1.14×10^(5) Da;红外光谱分析显示其具有糖的典型特征吸收峰。研究可为珊瑚菌液体培养菌丝体开发利用提供参考依据。Taking the liquid culture mycelium of Ramaria botrytoides as raw material,based on the single factor test,the extraction process of polysaccharide from Ramaria botrytoides mycelium was optimized by response surface methodology,and the structure of the extracted mycelium polysaccharide was preliminarily analyzed.The results showed that the optimum extraction conditions were as follows:solid-liquid ratio 1∶25(g/mL),extraction temperature 83℃ and extraction time 4.2 h.Under these conditions,the average yield of mycelium polysaccharide was(15.10±0.26)%.High performance liquid chromatography(HPLC)pre column derivatization analysis showed that it contained mannose,ribose,rhamnose,glucose,galactose,xylose and arabinose,and the corresponding molar ratio was 0.12∶0.55∶0.41∶1.00∶0.20∶1.15∶0.82.High performance gel permeation chromatography analysis showed that it mainly included two polysaccharide components,with relative molecular weights of 9.65×10^(5) Da and 1.14×10^(5) Da,respectively.Infrared spectrum analysis showed that there was a typical characteristic absorption peak of sugar.The study could provide a reference for the development and utilization of liquid culture mycelium of Ramaria botrytoides.
