摘 要:使用碱性蛋白酶对紫苏籽粕蛋白酶解,酶解物依次通过超滤和柱层析进行分离纯化,纯化肽经LC-MS-MS鉴定并通过动物实验对其进行降血脂及抗氧化功能性研究。结果表明,蛋白酶解物经超滤分离后,其分子质量小于3 ku的组分具有较高的抗氧化活性、该组分经DEAE-32离子交换色谱分离后,得到D1、D2、D3共3个组分,其中D3的抗氧化活性最好,D3经Sephadex G-25凝胶层析分离纯化后,富集得到G1、G2、G3三个组分,组分G2抗氧化活性最高。抗氧化活性肽G2对1,1-二苯基-2-三硝基苯肼(DPPH)、2,2′-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)、超氧阴离子(O_(2)^(-))·、羟基(·OH)自由基清除率及还原力依次为70.00%、47.09%、51.17%、70.04%、0.742;通过LC-MS-MS鉴定,抗氧化活性肽G2组分由7个氨基酸组成,分子质量为790.3755 u,其氨基酸序列为Ala-Ala-Asp-Cys-Arg-Ala-Lys。经低、中、高剂量组G2灌胃处理后的小鼠相比高脂模型组,小鼠体内的MDA(丙二醛)含量显著或极显著下降(P<0.05或P<0.01);小鼠的GSH-Px(谷胱甘肽过氧化物酶)、SOD(超氧化物歧化酶)和CAT(过氧化氢酶)活力显著或极显著升高(P<0.05或P<0.01);小鼠体内的TC(总胆固醇)、TG(甘油三酯)和LDL-C(低密度脂蛋白胆固醇)含量显著或极显著降低(P<0.05或P<0.01),HDL-C(高密度脂蛋白胆固醇)含量显著或极显著升高(P<0.05或P<0.01)。Alkaline proteases were usedfor enzymolysis of perila seed meal,and the zymolytes were separated and purified byultrafiltration and column chromatography.The purfied peptide was idenfified by,and the research on liqid lowering and antioxidantfunction were conducted by animal experiment.The results indicatedthat after the protease wasseparated by ultrafiltration,the component having a molecular weight of less than 3 ku hada high antioxidant activity,and three components of D1,D2,D3 were obtained by DEAE-32 ion exchange chromatography.Wherein,the antioxidant activity of D3 was the highested.After D2 was separated and purified by G-25 gel chromatography,three components of G1,G2,G3 was obtained by enrichment,and the component G2 has the highest antioxidant activity.The scavenging rates and reducing power of antioxidant peptide G2 to 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH),2,2′-diazo-bis-3-ethylbenzothiazoline-6-sulfonic acid(ABTS),superoxide anion(O_(2)^(-))and hydroxyl(·OH)radical were 70.00%,47.09%,51.17%and 70.04%,respectively,and the reducing power of G1 was 0.742.The antioxidant active peptide G2 components were composed of 7 amino acids by LC-MS-MS,and the molecular mass was 790.3755 u,and the amino acid sequence was Ala-Ala-ASP-CYS-ARG-ALA-ALAS.Compared with the high-fat model group,the content of MDA in mice treated by G2 gavage in low,middle and high dose groups decreased significantly or extremely significantly(P<0.05 or P<0.01);GSH-Px,SOD and CAT viability in mise was significanly or extremely significantly incaresed(P<0.05 or P<0.01);TC,Tg and LDL-C in mice significantly orextremely significantly decreased(P<0.05 or P<0.01),HDL-The C content was significantly orextremely significantly increased(P<0.05 or P<0.01).
