摘 要:为了简便快速地测定糕点中的富马酸二甲酯和纳他霉素,本文建立了一种以高效液相色谱同时测定糕点中的富马酸二甲酯和纳他霉素含量的分析方法。样品经75%甲醇超声提取后,采用Agilent TC-C色谱柱(150 mm×4.6 mm,5 μm)分离,以甲醇∶水为70∶30(V/V)为流动相,二极管阵列检测器检测,并用峰面积外标法来定量。方法采用了加标的方式来进行验证,分别对同一富马酸二甲酯和纳他霉素的加标样品进行了6次精密度试验。结果显示,富马酸二甲酯的回收率在97%~104%,相对标准偏差为0.71%~0.94%,检出限为0.004 μg·mL,定量限为0.01 μg·mL^(-1);纳他霉素的回收率在95.1%~99.3%,相对标准偏差为0.20%~0.86%,检出限为0.004 μg·mL^(-1),定量限为0.01 μg·mL^(-1)。结果表明,该方法的操作特点为简便、快速、准确,可同时测定糕点中富马酸二甲酯和纳他霉素含量的测定。In order to easily and quickly determine the dimethyl fumarate and natamycin in cake,an analytical method for the simultaneous determination of dimethyl fumarate and natamycin content in cake by high performance liquid chromatography is established.After ultrasonic extraction of 75% methanol,the samples were separated by Agilent TC-Ccolumns (150 mm×4.6 mm,5 μm),methanol∶water 70∶30(V/V) as the mobile phase,diode array detector detection,and quantified by peak area external standard method.Methods Were verified by means of standardization,and the standardization samples of the same dimethyl fumarate and natamycin were subjected to six precision experiments,and the recovery rate of dimethyl fumarate was between 97%~104%,the relative standard deviation was 0.71%~0.94%,the detection limit was 0.004 μg·mL^(-1),and the quantitative limit was 0.01 μg·mL^(-1).The recovery rate of natamycin ranged from 95.1% to 99.3%,the relative standard deviation was 0.20% to 0.86%,the detection limit was 0.004 μg·mL^(-1),and the quantitative limit was 0.01 μg·mL.The conclusion shows that the operation characteristics of this method are simple,fast and accurate,and the determination of dimethyl fumarate and natamycin content in cake can be determined simultaneously.
