摘 要:本文建立了高效液相色谱法测定食品添加剂玉米黄中特征成分玉米黄素含量的分析方法。样品用乙醇超声提取,样液经0.22μm滤膜过滤后进样分析。色谱柱:C30色谱柱(250mm×4.6mm,5μm);流动相:甲醇/乙腈(60+40,体积比),甲基叔丁基醚;洗脱方式:梯度,25min;检测器波长:446nm;流速:1.0mL/min;柱温:室温;进样量:10μL;采用外标法定量分析。结果表明,玉米黄素在0.5~10mg/L范围内与吸收度呈良好的线性关系(r≥0.9999),平均加标回收率在96.2%~105%之间,相对标准偏差(n=6)在1.53%~5.74%范围内,本方法测定玉米黄素快速准确、操作简单,重现性好,可用于商品化玉米黄的含量测定。In this study,a method for the determination of the characteristic component of food additive corn yellow by high performance liquid chromatography(HPLC)was established. The samples were extracted with ethanol,and extracted solution was filtered through a 0.22μm filter and then injected for analysis. The detection condition was :C30 column(250mm×4.6mm,5μm);mobile phase :methanol/acetonitrile(60+40,volume ratio),methyl tert-butyl ether;elution mode :gradient,25min;detector wavelength :455nm;flow rate : 1.0mL/min;column temperature :room temperature;injection volume :10μL;quantitative analysis performed by external standard method. The results showed that the linearly dependent coefficient is 0.9999 in the concentration range of 0.5 ~ 10mg/L,the average recovery rate was between 96.2% and 105%,the relative standard deviation(n=6)was in the range of 1.53% to 5.74%. With the advantages of rapidness,accuracy,simplicity,and good reproducibility,the method is suitable for determination zeaxanthin in food additive.
