摘 要:目的:建立高效液相色谱法测定红茶中茶黄素含量的分析方法。方法:样品用70%甲醇为提取溶液,在75℃水浴中提取15 min,提取2次,流动相为2%的乙酸水溶液和乙腈-乙酸乙酯(42∶3),波长280 nm,外标法定量。结果:方法线性良好(R^(2)>0.999),加标回收率为95.9%~98.2%,精密度良好(RSD≤4.7%)。对贵州喀斯特地区12个茶树品种红茶及其他地区10种红茶中的茶黄素含量进行测定,不同品种红茶中4种茶黄素的含量存在一定差异,贵州喀斯特地区红茶的茶黄素平均含量较其他地区要高。结论:贵州喀斯特地区对茶黄素总量有一定的影响。Objective:To establish an analytical method for the determination of theaflavin in black tea by high performance liquid chromatography(HPLC).Method:The sample was extracted twice with 70%methanol as the extraction solution in a 75℃water bath for 15 min.The mobile phase was 2%acetic acid aqueous solution and acetonitrile ethyl acetate(42:3),wavelength of 280 nm,and quantitative determination by external standard method.Result:The linearity of the method was good(R^(2)>0.999),the recovery was 95.9%~98.2%,and the precision was good(RSD≤4.7%).The contents of theaflavins in 12 tea varieties of black tea in karst areas of Guizhou and 10 kinds of black tea in other areas were determined.There were certain differences in the contents of 4 kinds of theaflavins in different varieties of black tea.The average content of theaflavins in black tea in karst area of Guizhou was higher than that in other areas.Conclusion:The total amount of theaflavins had a certain effect in karst areas of Guizhou.
