摘  要:以地皮菜糖蛋白提取得率为评价指标,利用单因素试验及Box-Behnken设计考察液料比、乙醇体积分数、提取温度、提取时间对地皮菜糖蛋白提取工艺的影响,对地皮菜糖蛋白进行硒化得到地皮菜硒化糖蛋白,并对二者开展体内抗氧化活性比较。结果表明,地皮菜糖蛋白最佳提取条件为:液料比11mL/g,乙醇体积分数62%,提取温度52℃,提取时间140min,此条件下地皮菜糖蛋白提取得率为(9.63±0.29)%。体内抗氧化试验结果显示,地皮菜硒化糖蛋白及地皮菜糖蛋白均能降低S180荷瘤小鼠血清和肝脏中丙二醛(MDA)的含量,升高过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活力和水平,且地皮菜硒化糖蛋白的抗氧化活性优于地皮菜糖蛋白。地皮菜糖蛋白及其硒化产物具有作为功能配料用于功能性食品开发的潜力。Taking the extraction yield of glycoprotein form Nostoc commune Vaucher as the evaluation index,the effects of liquid-solid ratio,ethanol volume fraction,extraction temperature and extraction time on the extraction process of glycoprotein from Nostoc commune Vaucher were investigated by single factor test and Box-Behnken design.The glycoprotein was reacted to Se-glycoprotein,and their antioxidant activities were compared in vivo.The results showed that the optimum extraction conditions of glycoprotein from Nostoc commune Vaucher were as follows:liquid-solid ratio 11mL/g,ethanol volume fraction 62%,extraction temperature 52℃,and extraction time 140min.Under these conditions,the extraction yield of glycoprotein from Nostoc commune Vaucher was(9.63±0.29)%.In addition,the results of antioxidant assays in vivo showed that Se-glycoprotein and glycoprotein from Nostoc commune Vaucher could reduce the content of malondialdehyde(MDA)in serum and liver of S180 tumor bearing mice,increase the activity and level of catalase(CAT),superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px),and the antioxidant activity of Se-glycoprotein was better than that of glycoprotein.The glycoprotein from Nostoc commune Vaucher and its selenide have the potential to be used as functional ingredient in the development of functional foods.
