摘 要:利用薄膜分散超声波法制备姜黄油脂质体,单因素实验考察了姜黄油体积分数、大豆卵磷脂和胆固醇比例、超声时间和超声功率对姜黄油包封率的影响,响应面法优化并获得了姜黄油脂质体的制备工艺为姜黄油体积分数1.2%,卵磷脂和胆固醇5∶1,超声时间21 min,此条件下姜黄油包封率可达到96%,建立的响应面模型对姜黄油脂质体的制备具有指导意义。性能分析表明,姜黄油脂质体的粒径为896 nm,PDI多分散指数为0.44,分别在4℃和25℃的条件下储存14 d内的pH均保持稳定。在10%乙醇和50%乙醇的模拟释放液中12 h内的姜黄油累计释放率分别为11.7%和21.5%,具有一定缓释效果,释放过程分别符合Ritger-peppas和一级释放动力学模型。姜黄油脂质体对单增李斯特菌具有长效的抑制作用,研究对于提高姜黄油的稳定性、实现长效抑菌效果具有重要意义。Turmeric oil liposome was prepared by the thin-membrane dispersion and sonication method.The single factor experiments included the turmeric oil volume concentration,the soy lecithin to cholesterol ratio,ultrasonic time and ultrasonic power.The optimum preparation process of turmeric oil was determined as 1.2%of turmeric oil volume concentration,5∶1 of lecithin to cholesterol,and 21 min of ultrasonic time.Under these conditions,the encapsulation rate of turmeric oil could reach 96%.The results indicated that,the size of turmeric liposomes was 896 nm,the PDI polydispersion index was 0.44,and the pH of turmeric oil liposome remained stable at 4℃and 25℃for 14 days,respectively.The cumulative release rates of turmeric oil in simulated release solution of 10%ethanol and 50%ethanol were 11.7%and 21.5%,respectively,with a certain sustained release effect.The release process complied with Ritger-Peppas and first-order release kinetics models,respectively.Furthermore,turmeric oil liposome had a long-term inhibitory effect on listeria monocytogenes.The study was of great significance to improve the stability of turmeric oil and achieve long-term bacteriostatic effect.
