摘  要:以刺玫果为原料,采用酶辅助水提法提取刺玫果多糖,以加酶量、酶解时间、料液比为考察因素,多糖提取率为评价指标,采用单因素试验和Box-Behnken响应面设计建立数学模型,应用遗传算法对模型进行优化,得到提取工艺最佳条件,并对刺玫果多糖进行抗氧化活性初步研究。结果表明:酶辅助法提取刺玫果多糖的最佳工艺条件为加酶量1.37%,酶解时间71min,料液比1∶30g/mL。Box-Behnken响应面法和遗传算法联用时,刺玫果多糖提取率高,为16.38%。刺玫果多糖具有显著的自由基清除作用,但与抗坏血酸对比,其对DPPH自由基、ABTS~+自由基和羟自由基的清除率均略低。Rosa davurica Pall was used as raw material,and polysaccharide was extracted by cellulase assisted water extraction method. The enzyme dosage,enzymatic hydrolysis time and solid-liquid ratio were selected as investigation factors and the polysaccharide extraction rate was the evaluation index. The mathematical model was established by single factor test and Box-Behnken response surface design. The model was optimized by genetic algorithm to obtain the best extraction conditions,and antioxidant activity of polysaccharide from Rosa davurica Pall was studied. The results showed that the optimum extraction conditions were as follows :enzyme dosage 1.37%,enzymatic hydrolysis time 71min,and solid-liquid ratio 1∶30g/mL. When Box-Behnken response surface design method was combined with genetic algorithm,the extraction rate of polysaccharide from Rosa davurica Pall was as high as 16.38%. The polysaccharide from Rosa davurica Pall had significant free radical scavenging effect. Compared with ascorbic acid,it had slightly lower scavenging rates of DPPH free radical,ABTSfree radical and hydroxyl free radical.
