摘 要:目的 优化烤鱼片类水产制品中淀粉含量的测定方法,探索提高该类产品中淀粉含量检测准确度和检测效率的方法。方法 在探索测定该类产品淀粉含量最佳条件时,通过改变乙醇浓度(75%、80%、85%)、乙醇洗涤量(150、250、300 mL)、振摇时间(10、20、30 min),设计国标法L_(9)(3^(4))正交试验,分析对淀粉含量测定结果的影响,并用3,5-二硝基水杨酸法(3,5-Dinitrosalicylic acid method,DNS法)进行测定,与国标法测定结果进行比较。结果 优化后除去可溶性糖的最佳条件:乙醇洗涤量为300 mL,乙醇浓度85%,振摇时间为20 min。DNS法回收率为91.54%~93.10%。结论 优化前处理方式,可以减少可溶性糖对结果的干扰,提高检测结果的准确度,比色法比国标法更高效、准确,可适用于该类产品的检测。Objective To optimize the determination method of starch content in grilled fish fillet aquatic products,explore methods to improve the detection accuracy and efficiency of starch content in such products.Methods When exploring the best conditions for determining the starch content of such products,the national standard method L_(9)(3^(4)) orthogonal test was designed to analyze the influence on the determination results of starch content by changing the conditions such as ethanol concentration(75%,80%,85%),ethanol washing amount(150,250,300 mL) and shaking time(10,20,30 min).The results were determined by 3,5-Dinitrosalicylic acid method(DNS) and compared with those by national standard method.Results The optimized conditions for removing soluble sugars were as follows:the washing volume of ethanol was 300 mL,the concentration of ethanol was 85%,and the shaking time was 20 min.The recovery rate of DNS method was 91.54%-93.10%.Conclusion The optimized pre-processing method can reduce the interference of soluble sugars on the results and improve the accuracy of the detection results,and the colorimetric method is more efficient and accurate than the national standard method,which can be applied to the detection of this kind of product.
