摘  要:以恩施富硒四季豆叶片为材料,采取碱提酸沉法提取富硒四季豆叶蛋白(leaf protein of selenium-enriched kidney bean,SKP).在单因素试验基础上,考察了提取时间、提取温度、料液比、碱液浓度4个因素,以SKP提取率作为响应值,采用响应面法优化SKP的提取工艺.结果表明,提取SKP的最优条件是:提取时间为120 min、提取温度为60℃、料液比为1∶20 g/mL、NaOH溶液浓度为0.2 mol/L;在此条件下,SKP的提取率为(29.92±1.28)%.氨基酸分析表明,该蛋白质中含有7种人体必需氨基酸.测定了SKP的体外抗氧化活性,在其质量浓度0.5~3.0 mg/mL范围内,DPPH自由基、ABTS自由基、羟基自由基、超氧阴离子自由基的清除率均超过50%,表明SKP具有良好的抗氧化能力.该结果为富硒四季豆的开发利用提供了一定的理论依据.In this paper,leaf protein of selenium-enriched kidney bean(SKP)was extracted from selenium-enriched kidney bean leaves in Enshi by alkali-extraction and acid-precipitation method.On the basis of single factor experiment,four factors,namely,extraction time,extraction temperature,solid-liquid ratio and lye concentration,were investigated,and the extraction rate of SKP was used as the response value.The response surface methodology was used to optimize the extraction process of SKP.The results showed that the optimal extraction conditions as follows:extraction time was 120 min,extraction temperature was 60℃,solid-liquid ratio was 1∶20 g/mL,NaOH concentration was 0.2 mol/L;under these conditions,the extraction rate of SKP was(29.92±1.28)%.Amino acid analysis shows that the protein contains 7 kinds of essential amino acids.The in vitro selenium-enriched kidney bean leaf protein was determined.In the range of selenium-enriched kidney bean leaf antioxidant activity of protein concentration of 0.5~3.0 mg/mL,DPPH radical,ABTS radical,hydroxyl radical and superoxide anion free radical clearance rates were all over 50%,indicating that the SKP had good antioxidant capacity.The results provide a theoretical basis for the development and utilization of selenium-enriched kidney beans.
