摘 要:为了研究制备目标天然产物分子溶致液晶模板以及对其进行分离纯化的工艺技术及机理,文章选择了苜蓿中的生物活性成分进行提取,研究使用乙酸乙酯、乙醇、水制备苜蓿醇提物中氨基酸H-Asp(OH)-OH溶致液晶模板,观察选择适宜的溶致液晶模板织构形式,在一定的条件下,对目标分子进行结晶纯化,并对获得的目标结晶物进行核磁共振检测证实结晶体为H-Asp(OH)-OH,同时在研究过程中观察、分析并提出了制备溶致液晶模板对天然产物分子进行分离纯化的机理假设——化学反应模板理论。制备溶致液晶模板分离纯化天然产物分子是一种具有高效性与靶向性的新型分离纯化技术,技术实施条件温和,极其适合具有生物活性物质的纯化分离,因而研究其工艺技术及机理极具研究价值。In order to study the technology and mechanism of separation and purification of the target natural product molecular lyotropic liquid crystal template,in this paper,the bioactive components in alfalfa were selected for extraction,the amino acid H-Asp(OH)-OH sol liquid crystal template in alfalfa ethanol extract was prepared with ethyl acetate,ethanol and water,and the appropriate texture form of sol liquid crystal template was observed and selected.Under certain conditions,the target molecule was crystallized and purified,and the obtained target crystal was confirmed to be H-Asp(OH)-OH by nuclear magnetic resonance detection.At the same time,the mechanism of isolation and purification of natural product molecules by preparing sol liquid crystal template,the theory of chemical reaction template was observed,analyzed and proposed in the process of research.The preparation of lyotropic liquid crystal template for the separation and purification of natural product molecules is a new separation and purification technology with high efficiency and targeting,the technical implementation conditions are mild,and it is extremely suitable for the purification and separation of bioactive substances.Therefore,the research on its process technology and mechanism is of great research value.
