摘 要:目的集成分子印迹技术、静电纺丝技术和微流控技术构筑分子印迹微流控芯片,设计能够承载纳米纤维膜的微流控芯片,并研究其在玉米赤霉烯酮前处理富集中的应用。方法以沉淀聚合法合成玉米赤霉烯酮分子印迹聚合物微粒,通过分子印迹材料与聚丙烯腈纺丝液的比例优化,筛选距离、电压、流速等静电纺丝条件,最终在纺丝距离15 cm、纺丝电压17 kV、流速控制30μL/min等纺丝条件下,制备了对目标物具有特异性识别的分子印迹纳米纤维膜;结合AutoCAD设计、雕刻机铣削加工等制膜条件,获得分子印迹微流控芯片,通过恒流泵将样品样液匀速通入芯片,利用芯片内部分子印迹纳米纤维膜进行富集。结果微流控分子印迹纳米膜对玉米赤霉烯酮及其代谢物的富集可达75.56%~94.50%,相对标准偏差在0.43%~2.66%。结论该芯片对目标物具有很好的吸附效果,能够应用于玉米中玉米赤霉烯酮的富集与净化。Objective To design a microfluidic chip capable of carrying nanofiber membranes,and study its application in the enrichment of zearalenone pretreatment.Integrate molecular imprinting technology,electrospinning technology and microfluidic technology to construct molecular imprinting microfluidic chip.Methods Zearalenone molecularly imprinted polymer particles were synthesized by precipitation polymerization.By optimizing the proportion of molecularly imprinted material and polyacrylonitrile spinning solution,screening the electrospinning conditions such as distance,voltage and flow rate.Finally,under spinning conditions such as spinning distance of 15 cm,spinning voltage of 17 kV and flow rate control of 30μL/min,molecularly imprinted nanofiber membrane with specific recognition of the target was prepared.Combined with AutoCAD design,engraving machine milling and other membrane preparation conditions,the molecule-imprinted microfluidic chip was obtained,the sample liquid was passed into the chip at a uniform speed by a constant current pump,and molecularly imprinted nanofiber membranes inside the chip for enrichment.Results The enrichment of microfluidic molecularly imprinted nanofilm on zearalenone and its metabolites could reach 75.56%‒94.50%,and the relative standard deviations were 0.43%‒2.66%.Conclusion The chip has good adsorption effect on the target material,and can be used for the enrichment and purification of zearalenone in maize.
