摘 要:目的探讨γ辐照结合钙调素拮抗剂三氟啦嗪(trifluoperazine,TFP)对冷藏蓝莓果实硬度品质特性的影响机制。方法采用2.5kGy剂量辐照处理“蓝丰”蓝莓,结合钙调素拮抗剂TFP测定各组蓝莓果实硬度、原果胶、不同溶解性果胶、纤维素和半纤维素含量,以及相关水解酶活性在冷藏期间的变化差异。结果辐照能够介导Ca^(2+)信号转导抑制多聚半乳糖醛酸酶(polygalacturonase,PG)和果胶甲酯酶(pectinmethylesterase,PME)活性,降低原果胶和螯合剂溶性果胶(chelator-soluble pectin,CSP)降解为水溶性果胶(water-soluble pectin,WSP)的可能性,进而对冷藏蓝莓果实硬度产生积极影响;TFP能够在一定程度上阻断胞内Ca^(2+)信号的转导作用;但纤维素酶(cellulase,Cx)、β-半乳糖苷酶(β-galactosidase,β-Gal)和α-L-阿拉伯呋喃糖苷酶(α-L-arabinofuranosidase,α-Af)对辐照诱导的Ca^(2+)信号响应不积极,辐照后冷藏蓝莓果实连接松散型半纤维素(4%KOH-soluble hemicellulose,4KSF)、连接紧密型半纤维素(24%KOH-soluble hemicellulose,24KSF)和纤维素含量的变化与Ca^(2+)信号转导无关。结论γ辐照可以介导Ca^(2+)信号通过抑制果胶降解对冷藏蓝莓果实硬度品质产生积极影响。Objective To investigate the effect mechanism ofγirradiation combined with calmodulin-antagonist triflurazine(TFP)on the hardness and quality characteristics of refrigerated blueberry fruits.Method“Bluecrop”blueberry fruits were treated byγirradiation at a dose of 2.5 kGy.Combining the treatments with calmodulinantagonists TFP,fruit firmness,content of protopectin and pectins of different solubilities,cellulose and hemicellulose content,and the changes of related hydrolase activities during cold storage in each group were detected.Resultsγirradiation could mediate the Ca^(2+) signal transduction to inhibit polygalacturonase(PG)and pectin methylesterase(PME)activities,reduced the probability of degradation of protopectin and chelator-soluble pectin(CSP)into water-soluble pectin(WSP).In this way,γirradiation could positively influence the firmness of refrigerated blueberry fruit,and TFP could inhibit the signal transduction to a certain degree.However,cellulase(Cx),β-galactosidase(β-Gal),andα-L-arabinofuranosidase(α-Af)responded inactively to the irradiation-induced Ca^(2+) signals,and changes in 4%KOH-soluble hemicellulose(4KSF),24%KOH-soluble hemicellulose(24KSF),and cellulose content had little influence on Ca^(2+) signal transduction.Conclusionγirradiation can mediate Ca^(2+) signal and have a positive impact on the firmness and quality of blueberry fruits during cold storage by inhibiting pectin degradation.
