摘 要:为探究长茎葡萄蕨藻醇提物不同溶剂(石油醚、乙酸乙酯、正丁醇、水)萃取物对胰脂肪酶及α-葡萄糖苷酶的抑制作用,该试验采用比色法测定长茎葡萄蕨藻醇提物不同极性溶剂萃取物中多酚、黄酮、萜类的含量,酶底物反应法测定对胰脂肪酶及α-葡萄糖苷酶的抑制活性,并研究二者的相关性。采用酶抑制动力学方法和荧光光谱技术研究抑制作用最强的萃取物对胰脂肪酶及α-葡萄糖苷酶的互作机制。结果表明:长茎葡萄蕨藻醇提物不同溶剂萃取物中多酚(0.62 mg/g~23.79 mg/g)、萜类(6.50 mg/g~214.29 mg/g)、黄酮(0.35 mg/g~3.27 mg/g)的含量均具有差异性。醇提物的不同溶剂萃取物抑制胰脂肪酶活性强弱为乙酸乙酯萃取物>石油醚萃取物>水萃取物>正丁醇萃取物,抑制α-葡萄糖苷酶活性强弱为乙酸乙酯萃取物>正丁醇萃取物>石油醚萃取物>水萃取物。长茎葡萄蕨藻醇提物乙酸乙酯萃取物对胰脂肪酶和α-葡萄糖苷酶的抑制作用最强(IC50分别为1.662 mg/g和0.017 mg/mL)。相关性分析表明提取物的萜类和多酚含量与胰脂肪酶抑制活性呈显著相关(p<0.05),多酚含量与α-葡萄糖苷酶抑制活性呈显著相关(p<0.05)。酶抑制动力学研究表明,长茎葡萄蕨藻醇提物乙酸乙酯萃取物对胰脂肪酶的抑制类型为非竞争性抑制,对α-葡萄糖苷酶的抑制类型为反竞争性抑制。荧光光谱分析表明,长茎葡萄蕨藻醇提物乙酸乙酯萃取物对胰脂肪酶和α-葡萄糖苷酶的猝灭方式均为静态猝灭。长茎葡萄蕨藻醇提物的乙酸乙酯萃取物对胰脂肪酶与α-葡萄糖苷酶活性的抑制作用较强,可能与其中含有多酚与萜类物质有关。This study aimed to investigate the inhibitory effects of Caulerpa lentillifera alcohol extracts by different solvents(petroleum ether,ethyl acetate,n-butanol,and water)on pancreatic lipase and α-glucosidase.The content of polyphenols,flavonoids,and terpenoids in alcohol extracts of C. lentillifera was determined by colorimetry,and the inhibitory activities against pancreatic lipase and α-glucosidase were determined by enzyme substrate reaction. The correlations between them were studied. The interaction mechanisms of the extracts possessing the strongest inhibitory activity against pancreatic lipase and α-glucosidase were explored by enzyme inhibition kinetics and fluorescence spectroscopy. The results showed that the content of polyphenols(0.62 mg/g-23.79 mg/g),terpenoids(6.50 mg/g-214.29 mg/g),and flavonoids(0.35 mg/g-3.27 mg/g)in C. lentillifera alcohol extracts by different solvents were different. In terms of the inhibitory activity against pancreatic lipase,solvent extracts were ranked as ethyl acetate extract > petroleum ether extract > water extract> n-butanol extract. In terms of the inhibitory activity against α-glucosidase,solvent extracts were ranked as ethyl acetate extract > n-butanol extract > petroleum ether extract > water extract. Among them,C. lentillifera alcohol extract by ethyl acetate showed the strongest inhibition of pancreatic lipase and α-glucosidase with IC50values of 1.662 mg/g and 0.017 mg/mL,respectively. The content of triterpenoids and polyphenols was correlated with inhibitory activity against pancreatic lipase(p<0.05),and the content of polyphenols was correlated with the inhibitory activity against α-glucosidase(p<0.05). The enzyme inhibition kinetics showed that the C. lentillifera alcohol extract by ethyl acetate showed a non-competitive inhibition against pancreatic lipase and an anticompetitive inhibition against α-glucosidase. The fluorescence spectroscopy showed that the C. lentillifera alcohol extract by ethyl acetate showed static quenching of pancreatic lipase
