摘 要:采用单因素试验及响应面试验对纳豆芽孢杆菌(Bacillus natto)培养条件进行优化。在单因素试验的基础上,取影响纳豆激酶活力的5个重要条件因子(培养温度、培养时间、初始pH、接种量、菌龄)设计5因素3水平的响应面试验,建立以纳豆激酶活力为响应值的多元二次回归方程。结果表明,纳豆芽孢杆菌最佳培养条件为:培养温度34℃、培养基初始pH值为7.5、接种量3%、培养时间95 h、菌龄17.5 h。在此最佳条件下,纳豆激酶活力达到5087 FU/mL,较优化前提高了18.83%。The culture conditions of Bacillus natto were optimized by single factor tests and response surface tests.Based on single factor tests,5 important condition factors(culture temperature,time,initial pH,inoculum,cell age)that affecting the nattokinase activity were selected to design the response surface test with 5 factors and 3 levels,and the multiple quadratic regression equation with the activity of nattokinase as the response value was established.The results showed that the optimal culture conditions of B.natto were as follows:culture temperature 34℃,medium initial pH 7.5,inoculum 3%,culture time 95 h,cell age 17.5 h.Under the optimum conditions,the nattokinase activity reached 5087 FU/ml,which was 18.83%higher than that before optimization.
