摘  要:以海参性腺蛋白为研究对象,通过紫外光谱、傅里叶变换红外光谱和荧光光谱分析,表面疏水性和粒径分布测定,以及微观结构观察,对比不同时间(0、5、15、25、35 min,200 W)超声预处理对海参性腺蛋白中性蛋白酶酶解物结构特性的影响。结果表明:随着超声时间的延长,海参性腺蛋白酶解物的紫外吸光度、β-折叠相对含量、荧光强度、表面疏水性和粒径均呈现先升高后降低的趋势,α-螺旋相对含量的变化趋势与之相反。其中,当超声时间为15 min时,酶解物的紫外吸光度、β-折叠相对含量、荧光强度和表面疏水性分别较未超声时增加了7.30%、50.72%、4.50%和13.42%,α-螺旋相对含量降低了9.98%(P<0.05)。超声预处理15 min的酶解物具有最小的粒径尺寸和分布范围,此现象可以通过扫描电子显微镜观察结果更直观地展现。综上,适当的超声预处理可以促进海参性腺蛋白结构的展开,减少蛋白质的聚集,15 min超声预处理效果最佳。The effects of different periods(0,5,15,25 and 35 min)of ultrasound pretreatment at a power of 200 W on the structural characteristics of sea cucumber gonad protein hydrolysates prepared with neutral protease were investigated by ultraviolet-visible(UV-Vis)spectroscopy,Fourier transform infrared(FTIR)spectroscopy and fluorescence spectroscopy as well as determination of surface hydrophobicity and particle size distribution and microstructure observation.The results showed that the UV absorbance,β-sheet relative content,fluorescence intensity(FI),surface hydrophobicity and particle size of sea cucumber gonad protein hydrolysates initially increased and then decreased with ultrasound pretreatment time,while the opposite trend was observed forα-helix relative content.Ultrasound pretreatment for 15 min increased the UV absorbance,β-sheet relative content,FI and surface hydrophobicity of hydrolysates by 7.30%,50.72%,4.50%and 13.42%,respectively,and decreased theα-helix relative content by 9.98%(P<0.05).Moreover,this treatment resulted in the smallest particle size and distribution range of hydrolysates as observed under scanning electron microscopy(SEM).Therefore,appropriate ultrasound pretreatment could promote the unfolding of sea cucumber gonad protein structure and reduce protein aggregation,and the effect of ultrasound pretreatment for 15 min was most pronounced.
