摘 要:研究以斑马鱼高尿酸模型为试验对象,探讨经超微粉碎后的汉麻仁超临界脱脂粉的辅助降尿酸作用及其可能机制。采用10 mmol/L尿酸酶抑制剂氧嗪酸钾和0.5 mmol/L尿酸合成前体黄嘌呤钠盐共同处理受精后5 d (5 day post-fertilization, 5 dpf)的斑马鱼20 h,建立斑马鱼高尿酸模型。以别嘌呤醇为阳性对照,对高尿酸斑马鱼分别进行500, 1 000和2 000μg/mL这3种不同浓度汉麻仁脱脂粉水溶液浸泡处理,测定斑马鱼对汉麻仁脱脂粉的最大耐受浓度(maximum tolerance concentration, MTC)以及斑马鱼匀浆中的尿酸水平、次黄嘌呤鸟嘌呤-磷酸核糖转移酶(hypoxanthine phosphoribosyl transferase 1, HPRT1)和有机阴离子转运蛋白1 (organic anion transporter 1, OAT1)的mRNA表达。汉麻仁脱脂粉对模型斑马鱼的MTC为2 000μg/mL。与模型对照组相比,汉麻仁脱脂粉可通过上调斑马鱼匀浆中HPRT1相对表达量(P<0.05),有效降低尿酸水平(P<0.05),而对OAT1的相对表达量无显著差异(P>0.05)。汉麻仁脱脂粉具有辅助降尿酸功效,可能通过促进嘌呤的转化,抑制尿酸的产生,有望成为治疗高尿酸血症的潜在药物。This study was aimed to assess uric acid-lowering effect and its possible mechanisms of supercritical hemp seed defatted flour after superfine grinding in a zebra fish model. The zebra fish high uric acid model was constructed by feeding 5days post-fertilization zebra fish for co-treatment of uricase inhibitor potassium oxonate at 10 mmol/L and uric acid synthesis precursor xanthine sodium salt at 0.5 mmol/L for 20 h. Hemp seed defatted flour was administered to the high uric acid zebra fish through soaking at 500, 1 000 and 2 000 μg/ml, with allopurinol as a positive control. Maximum tolerance concentration(MTC)of hemp seed defatted flour for zebra fish, uric acid level, and mRNA expression of hypoxanthine guanine-phosphoribosyltransferases transferase(HPRT1) and organic anion transporter 1(OAT1) in whole juvenile zebra fish homogenate were measured.The MTC of the hemp seed defatted flour in fish water was 2 000 μg/mL in model zebra fish. Compared with control group,hemp seed defatted flour significantly reduced the level of uric acid through up-regulating HPRT1(P<0.05), but OAT1 gene expression was not significant(P>0.05). These results demonstrated that hemp seed defatted flour hold a potential for high uric acid prevention and therapy, possibly through promoting purine conversion and inhibiting uric acid production.
