摘 要:【目的】利用单因素结合响应面的研究方法,旨在使用真空冷冻干燥技术得到植物乳杆菌V02直投式乳酸菌发酵剂菌粉。【方法】以植物乳杆菌V02为试验菌种,冻干存活率为衡量指标,通过单因素试验结合Box-Behnken响应面法对不同保护剂的种类及质量分数进行筛选和复配,得到优化后的复配保护剂质量分数配比,最后进行验证试验。【结果】当选用质量分数分别为12.23%的脱脂乳粉,9.00%的麦芽糊精和6.50%的D-海藻糖时,复配保护剂对植物乳杆菌V02的冻干保护率可达到99.88%,相比空白组的冻干存活率提高了90.51%。【结论】经响应面优化后的植物乳杆菌V02保护剂配方可有效保护菌体在真空冷冻干燥过程中的菌体存活,此配方对于植物乳杆菌V02有较好的保护效果。[Objective]Single factor combined with response surface method was used to obtain Lactiplantibacillus plantarum V02 direct Lactobacillus starter culture powder by vacuum freeze-drying technology.[Methods]Lactiplantibacillus plantarum V02 was used as the experimental strain, and the lyophilized survival rate was used as the measurement index. The type and mass fraction of different protective age nts were screened and mixed by single factor experiment combined with Box-Behnken response surface method, and the optimized mass fraction ratio of the compound protective agent was obtained.[Results] When the mass fraction of skim milk powder(12.23%), maltodextrin(9.00%) and D-trehalose(6.50%) were used, the lyophilized protection rate of the compound protectant against Lactiplantibacillus plantarum V02 was 99.88%, which was 90.51% higher than that of the blank group.[Conclusion] The formula of Lactiplantibacillus plantarum V02 protective agent optimized by response surface can effectively protect the lyophilized survival rate of bacteria in vacuum freeze-drying process, and this formula has a good protective effect on Lactiplantibacillus plantarum V02.
