摘 要:以金钱白花蛇为原料,在单因素试验基础上采用响应面法优化其醇溶蛋白提取工艺,并进行十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)、高效液相色谱(HPLC)、傅里叶变换红外光谱(FTIR)指纹图谱分析。结果表明:最佳提取工艺条件为乙醇体积分数40%、料液比1∶17(g/mL)、提取时间4 h、提取温度40℃,此时醇溶蛋白得率为0.51%。SDS-PAGE指纹图谱分析表明金钱白花蛇醇溶蛋白分子质量主要介于26.69~88.02 kDa,26.69~28.34 kDa和70.84~88.02 kDa处的蛋白质丰度较高,分别占蛋白质总量的55.2%和31.8%;HPLC结果表明,主要金钱白花蛇醇溶蛋白的保留时间为2.928和3.452 min;FTIR结果表明,金钱白花蛇醇溶蛋白在3526.67、2918.31、2357.83、1940.24、1354.05、841.90、546.07和444.52 cm^(-1)处有特征吸收峰。试验结果可为金钱白花蛇醇溶蛋白资源的开发利用及药理活性研究提供依据。Using the Bungarus multicinctus as raw material,the extraction process of gliadin was optimized by response surface method on the basis of single factor test,and sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE),high performance liquid chromatography(HPLC)and Fourier transform infrared spectroscopy(FTIR)fingerprint analysis were carried out.The results showed that the optimum extraction conditions were as follows:ethanol volume fraction 40%,solid liquid ratio 1∶17(g/mL),extraction time 4 h and extraction temperature 40℃,and the yield ofgliadin was 0.51%.SDS-PAGE fingerprint analysis showed that the molecular weight of gliadin of the Bungarus multicinctus was mainly between 26.69-88.02 kDa,and protein abundance at 26.69-28.34 kDa and 70.84-88.02 kDa were higher,accounting for 55.2%and 31.8%of total protein,respectively.The results of HPLC showed that the retention time of the main gliadin of Bungarus multicinctus was 2.928 and 3.452 min.The results of FTIR showed that there were characteristic absorption peaks at 3526.67,2918.31,2357.83,1940.24,1354.05,841.90,546.07 and 444.52 cm^(-1).The results of this experiment can provide a basis for the development and utilization of the gliadin resources and the study of pharmacological activities of the Bungarus multicinctus.
