摘  要:目的 研究钝药野木瓜果实中果肉、果皮及种子的抗氧化及降血糖活性。方法 采用DPPH和ABTS自由基清除实验,铁还原力法检测其抗氧化活性,均以维生素C作阳性对照;采用α-葡萄糖苷酶及α-淀粉酶抑制模型评价其降血糖活性,均以阿卡波糖为阳性对照。结果 果皮清除DPPH自由基能力[IC_(50)=(0.586±0.016)mg/mL]和ABTS^(+)能力[IC_(50)=(2.850±0.08)mg/mL]显著强于果肉[IC_(50)=(1.177±0.029)mg/mL,(3.403±0.11)mg/mL]和种子[IC_(50)=(4.603±0.281)mg/mL,(15.984±0.19)mg/mL],果肉的铁还原能力[(164.530±3.78)μmol VitC/g]显著强于果皮[(147.330±1.51)μmol VitC/g]和种子[(75.010±3.73)μmol VitC/g](P<0.05)。相关性分析结果显示,钝药野木瓜果皮、种子和果肉的总酚、总黄酮含量与清除ABTS^(+)、DPPH自由基能力及铁还原能力均呈显著正相关(P<0.05)。果肉抑制α-葡萄糖苷酶活性[IC_(50)=(15.85±0.70)mg/mL]及α-淀粉酶活性[IC_(50)=(20.35±0.021)mg/mL]的能力显著强于果皮[IC_(50)=(30.16±1.30)mg/mL,(37.11±1.12)mg/mL]和种子[IC_(50)=(119.89±0.98)mg/mL,未测出(P<0.05)。结论 钝药野木瓜果实中果肉和果皮是天然抗氧化剂的潜在来源,同时钝药野木瓜果肉也具有成为降血糖药物的潜力。Objective To study the antioxidant activity and hypoglycemic activity of the pulp,peel and seeds in the fruit of Stauntonia leucantha.Methods DPPH and ABTS free radical scavenging tests and iron reduction method were adopted to detect its antioxidant activity with vitamin C as the positive control.The α-glucosidase and α-amylase inhibition models were adopted to evaluate the hypoglycemic effect of the fruit of Stauntonia leucantha with acarbose as the positive control.Results The DPPH free radical scavenging ability[IC_(50)=(0.586±0.016)mg/mL]and ABTS^(+)radical scavenging ability[IC_(50)=(2.850±0.081)mg/mL]of the peel were significantly better than those of the pulp[IC_(50)=(1.177±0.029)mg/mL,(3.403±0.11)mg/mL]and those of the seed[IC_(50)=(4.603±0.281)mg/mL,(15.984±0.19)mg/mL].The iron reduction ability of the pulp[(164.530±3.78)μmol VitC/g]was significantly better than that of the peel[(147.330±1.51)μmol VitC/g]and that of the seed[(75.010±3.73)μmol VitC/g](P<0.05).The correlation analysis result showed that the contents of total polyphenols and total flavonoids in the peel,seed and pulp of the fruit of Stauntonia leucantha were significantly positively correlated with the ABTS^(+)free radical scavenging ability,the DPPH free radical scavenging ability and the iron reduction ability (P<0.05).The α-glucosidase inhibition activity[IC_(50)=(15.85±0.70)mg/mL]and α-amylase inhibition activity[IC_(50)=(20.35±0.021)mg/mL]of the pulp were significantly better than those of the peel[IC_(50)=(30.16±1.30)mg/mL,(37.11±1.12)mg/mL]and those of the seed[IC_(50)=(119.89±0.98)mg/mL,not available](P<0.05).Conclusion The antioxidant activity result showed that the pulp and peel of the fruit of Stauntonia leucantha were potential sources of natural antioxidants.The hypoglycemic activity results showed that the pulp of Stauntonia leucantha has the potential to be developed into hypoglycemic drugs.
