摘 要:采用乙醇-硫酸铵双水相制备虾壳中的虾青素,探索虾壳中虾青素提取的条件。结果表明:采用乙醇溶液,料液比1∶15,每次提取3 h,提取2次,提取效果较好。采用乙醇-硫酸铵双水相萃取虾青素,虾青素主要集中在上下两相之间,最佳条件:20%硫酸铵∶虾青素乙醇提取液(v/v)=2.5∶2.0,沉淀率为89.09%。经薄层色谱法和高效液相色谱法检测,双水相萃取可使虾青素提取液中的各类型虾青素都沉淀。Astaxanthin in shrimp shell was prepared by ethanol/(NH 4)2SO 4 aqueous two-phase method.The extraction conditions of astaxanthin from shrimp shell were explored.It was determined that ethanol solution was used,the solid-liquid ratio was 1∶15,extracted for 3h and twice,and the extraction effect was better.Astaxanthin was extracted by ethanol/(NH 4)2SO 4 aqueous two-phase extraction.Astaxanthin was mainly concentrated between the upper and lower phases.The optimum condition was that 20%(NH 4)2SO 4∶astaxanthin ethanol extract(v/v)=2.5∶2.0,and the precipitation rate was 89.09%.By TLC and HPLC examination,all kinds of astaxanthin in astaxanthin extract can be precipitated by aqueous two-phase extraction.This environmentally friendly method has potential application prospects in the large-scale separation of astaxanthin.
