摘 要:为探究水产蛋白抗菌肽(Antibacterial peptide)作为天然食品保鲜剂的安全性,通过急性经口毒性试验、蓄积性毒性试验和小鼠精子畸形试验对海蟹源抗菌肽(Crab antibacterial peptide,CAP)、牡蛎源抗菌肽(Oyster antibacterial peptide,OAP)、带鱼源抗菌肽(Hairtail antibacterial peptide,HAP)和鲣鱼源抗菌肽(Bonito antibacterial peptide,BAP)4种水产蛋白抗菌肽进行了安全性评价。结果表明,海蟹源抗菌肽(CAP)、牡蛎源抗菌肽(OAP)、带鱼源抗菌肽(HAP)和鲣鱼源抗菌肽(BAP)对小鼠的急性经口毒性LD_(50)>10 g/kg.bw,属于实际无毒级。在剂量递增灌胃20 d后小鼠也未出现不良反应,属轻微蓄积毒性,且停药7 d后,小鼠体重和主要内脏质量均未见显著差异,心、肝、脾、肾等主要内脏外观颜色、大小均正常,未见明显病变,说明无明显的蓄积毒性。此外,海蟹源抗菌肽(CAP)、牡蛎源抗菌肽(OAP)、带鱼源抗菌肽(HAP)和鲣鱼源抗菌肽(BAP)对小鼠精子畸变率没有产生明显影响,未见生殖毒性。该结果为海蟹源抗菌肽(CAP)、牡蛎源抗菌肽(OAP)、带鱼源抗菌肽(HAP)和鲣鱼源抗菌肽(BAP)作为天然食品保鲜剂的安全性提供了科学依据。In order to explore the safety of aquatic protein antibacterial peptide as a natural food preservative. The acute oral toxicity test, cumulative toxicity test and mouse sperm deformity test were used to evaluate the safety of four aquatic protein antimicrobial peptides, including crab antibacterial peptide(CAP), oyster antibacterial peptide(OAP), hairtail antibacterial peptide(HAP) and bonito antibacterial peptide(BAP). The experimental results show that the acute oral toxicity LD_(50)>10 g/kg. bw of crab antibacterial peptide(CAP), oyster antibacterial peptide(OAP), hairtail antibacterial peptide(HAP)and bonito antibacterial peptide(BAP) on mice belongs to the actual non-toxic level. There was no adverse reaction in the mice after 20 days of dose increasing gavage, which was a slight cumulative toxicity. After 7 days of withdrawal, there was no significant difference in the body weight and main visceral weight of the mice. The appearance color and size of the main viscera such as heart, liver, spleen and kidney were normal, and no obvious lesions were observed, indicating no obvious cumulative toxicity. In addition,crab antibacterial peptide(CAP), oyster antibacterial peptide(OAP), hairtail antibacterial peptide(HAP)and bonito antibacterial peptide(BAP) had no significant effect on sperm aberration rate in mice, and no reproductive toxicity was observed. This experiment laid a scientific basis for the safety of crab antibacterial peptide(CAP), oyster antibacterial peptide(OAP), hairtail antibacterial peptide(HAP) and bonito antibacterial peptide(BAP) as natural preservatives for food.
