摘  要:以热风干燥和真空冷冻干燥2种方法制备广叶绣球菌子实体干制品,对2种干制品中可溶性多糖主要成分的分子量分布特性和体外抗炎活性进行比较分析。通过CCK-8法检测这2种可溶性多糖成分对小鼠腹腔巨噬细胞增殖的影响,并应用脂多糖(LPS)建立小鼠巨噬细胞RAW264.7炎症模型,考察细胞上清液中NO浓度及炎症因子TNF-α、IL-1β、IL-6浓度等指标,比较这2种绣球菌可溶性多糖成分的抗炎活性。结果表明,热风干燥品中可溶性多糖的主要成分SCG-G的重均分子量为1.48×104Da,真空冷冻干燥品中可溶性糖的主要成分SCG-D的重均分子量为2.47×105Da。绣球菌冻干品可溶性多糖中的主要成分SCG-D对小鼠巨噬细胞增殖活性影响较小,细胞毒性低,并能显著抑制LPS诱导的NO及炎症因子TNF-α、IL-1β、IL-6释放的作用,存在明显剂量效应关系,表现出较好的体外抗炎活性。There were two kinds of polysaccharide of Sparassis latifolia,one was SCG-G,which was isolated and purified from the hot air dried fruiting bodies,and the other was SCG-D,which was isolated and purified from vacuum freeze-drying dried fruiting bodies.The CCK-8 method was used to examine the effect of SCG-D and SCG-G on the proliferation of mouse peritioneal RAW264.7 macrophages in vitro under different concentrations.The production of NO,TNF-α,IL-1β,IL-6 were detected for inhibitory effect on LPS-induced inflammation in RAW264.7 cells in vitro.The results indicated that the molecular weight of SCG-G was 1.48×10~4 Da and the molecular weight of SCG-D was 2.47×10~5 Da.Furthermore,SCG-D exhibit lower level cytotoxicity with lower inhibiting effect on proliferation activity.Compared with the LPS-induced model,SCG-D showed superior inhibitory effect on NO,TNF-α,IL-1β,IL-6 production,with dose-response relationship visibly.
