摘 要:探索酶解法提取葡萄叶中白藜芦醇的最佳工艺,对农业废弃物的高效转化具有重要贡献。以葡萄叶为原料,采用酶解法提取白藜芦醇,并利用单因素及正交试验优化从葡萄叶中提取白藜芦醇的最佳工艺和研究提取物抑菌活性。结果表明:(1)酶解法提取葡萄叶白藜芦醇的最佳工艺条件为酶添加量0.8%、提取时间6 h、提取温度45℃、料液比1∶15,提取率最高达2.65%。(2)采用滤纸片法测定葡萄叶白藜芦醇的抑菌作用,发现当白藜芦醇浓度为41×10^(-2)mg/mL时对金黄色葡萄球菌、产气肠杆菌、变形杆菌、枯草芽孢杆菌及沙门氏杆菌均具有较好的抑菌效果;通过最低抑菌浓度MIC测定,发现葡萄叶白藜芦醇提取液对金黄色葡萄球菌的MIC为41×10^(-5)mg/mL,对产气肠杆菌的MIC为41×10^(-4)mg/mL,对变形杆菌和枯草芽孢杆菌的MIC为41×10^(-2)mg/mL,而对沙门氏杆菌的MIC为41×10^(-1)mg/mL。最终确定葡萄叶白藜芦醇对5种细菌的抑菌作用大小依次为:金黄色葡萄球菌>产气肠杆菌>变形杆菌=枯草芽孢杆菌>沙门氏杆菌。酶解法可以有效地提取葡萄叶白藜芦醇,葡萄叶白藜芦醇具有较好的抑菌作用,在天然抑菌剂的开发及应用方面具有广阔的发展前景。To explore the optimal extraction technology of resveratrols from Vitisvinifera leaves by enzymolysis,it has important contribution to transform agricultural waste efficiently.Using Vitisvinifera leaves as raw materials,the extraction process of resveratrols from grape leaves was optimized by single factor experiment and orthogonal design test,and its antibacterial activity is determined by filtering paper method.These results showed that the optimal extraction conditions as follows:enzyme additive amount is 0.8%,extraction time is 6h,extraction temperature is 45℃,solid-liquid ratio is 1∶15,and the extraction yield of resveratrols from Vitisvinifera leaves is 2.65%.The antibacterial activity of resveratrols is determined by filtering paper method.It is found that resveratrols have obvious inhibitory effect on Staphylococcus aureus Enteroaerogen,Proteus,Bacillus subtilis,and Salmonella at 41×10^(-2) mg/mL.Then,it is also found that resveratrols have minimal inhibitory concentration on Staphylococcus aureus at 41×10^(-5) mg/mL,it have MIC on Enterobacteraeriformis at 41×10^(-4) mg/mL,it have MIC on Bacillus subtilis and Proteus at 41×10^(-2) mg/mL,respectively,and it have MIC on Salmonella at 41×10^(-1) mg/mL.The antibacterial activity of resveratrol on 5 kinds of bacteria was as follows:Staphylococcus aureus>Enterobacteraerogenes>Proteus=Bacillus subtilis>Salmonella.The resveratrols from Vitisvinifera leaves can be effectively extracted by enzymolysis method.It has antibacterial effects and broad prospects in the application of natural antibacterial agents.
