摘 要:采用传统培养分离法从新疆发酵酸奶中分离乳酸菌菌株,通过钙富集量、耐受性与黏附能力测定筛选性能优良的富集钙菌株,通过抗生素敏感性、溶血活性与硝基还原酶产生能力测定进行安全性评价,并采用形态观察及分子生物学技术进行菌种鉴定。结果表明,共分离出40株乳酸菌,均具有钙富集能力,其中菌株M5、M18、M24的钙富集能力较好,分别为41.90 mg/g、49.37 mg/g、52.60 mg/g,且对酸和胆盐具有耐受性;菌株M24在氯仿中疏水性高(44.12%),菌株M18在二甲苯中疏水性高(37.14%),菌株M5、M24的自凝聚能力均>80%;3株菌株经人工胃肠液消化,活菌数均>6×10^(7)CFU/mL;3株菌株对氯霉素、克林霉素、氨苄西林、红霉素敏感,溶血试验阴性且不产生硝基还原酶,为安全菌株。经鉴定,菌株M5、M18、M24分别为发酵乳杆菌(Lactobacillus fermentum)、乳酸片球菌(Pediococcus acidilactici)和植物乳杆菌(Lactiplantibacillus plantarum)。Lactic acid bacteria were isolated from fermented yogurt in Xinjiang by traditional culture separation method,the calcium-enriched strains with excellent performance were screened by the determination of calcium concentration,tolerance and adhesion ability,the safety evaluation was carried out by determining of antibiotic sensitivity,hemolytic activity and nitroreductase production ability,and the strains were identified by morphological observation and molecular biology techniques.The results showed that 40 strains of lactic acid bacteria were isolated,all strains had calcium enrichment ability,among which the strains M5,M18 and M24 had better calcium enrichment ability,which were 41.90 mg/g,49.37 mg/g and 52.60 mg/g,respectively,and were tolerant to acid and bile salts.The strain M24 had high hydrophobicity in chloroform(44.12%),and the strain M18 had high hydrophobicity in xylene(37.14%).The autoagglutination ability of strains M5 and M24 was greater than 80%.The viable bacterial counts of the 3 strains were greater than 6×10^(7) CFU/ml after digestion by artificial gastroenteric fluid.The 3 strains were sensitive to chloramphenicol,clindamycin,ampicillin and erythromycin,the hemolysis test was negative and didn't produce nitroreductase,so they were safe.The strains M5,M18 and M24 were identified as Lactobacillus fermentum,Pediococcus acidilactici,and Lactiplantibacillus plantarum.
