摘  要:目的通过研究苦丁茶冬青高效液相色谱法(HPLC)指纹图谱与抗氧化活性的关系,初步确定其抗氧化活性物质基础,为其质量控制方法研究提供依据。方法构建多酚类化合物的HPLC指纹图谱,运用中药色谱指纹图谱相似度评价系统(2012版)确定共有峰,运用SPSS22.0软件进行主成分分析。1,1-二苯基-2-苦基肼(DPPH)法和铁氰化钾法测定不同产地的苦丁茶冬青抗氧化活性,采用Pearson、Spearman双变量相关分析和偏最小二乘回归分析苦丁茶冬青多酚类化合物与抗氧化活性的谱效关系。HPLC测定通过谱效学研究确认的化合物含量,并进行方法学考察。结果由主成分分析可知新绿原酸、隐绿原酸、峰9在苦丁茶冬青样品区分中有较大影响。新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、异绿原酸C、咖啡酸、异槲皮素具有较好的抗氧化活性。聚类分析表明不同产地的苦丁茶冬青样品呈现出一定的地域性聚类特征。含量测定方法学良好。结论通过将苦丁茶冬青指纹图谱与抗氧化活性进行整合分析,确定了苦丁茶冬青中各成分的抗氧化活性,为阐明苦丁茶冬青中抗氧化活性物质基础及质量控制方法制定提供实验依据。OBJECTIVE To determine the material basis of antioxidant activityof Ilex kudingcha through the study of the relationship between the HPLC fingerprint and antioxidant activity of Ilex kudingcha and provide the basis for the research of quality control method.METHODS The HPLC fingerprint of polyphenolic compounds was constructed,and the common peaks were determined by the TCM chromatographic fingerprint similarity evaluation system(2012 edition),and the principal component analysis was performed by SPSS 22.0 software.DPPH method and potassium ferricyanide method were used to determine the antioxidant activity of different producing areas of Ilex kudingcha,Pearson,Spearman bivariate correlation analysis and partial least squares regression analysis were used to analyze the relationship between polyphenolic compounds and antioxidant activity of Ilex kudingcha.HPLC method was used to determine the content of compounds confirmed by spectral effect study,and methodological investigation was conducted.RESULTS According to the principal component analysis,neochlorogenic acid,cryptochlorogenic acid and peak 9(not confirmed)had a great influence on the identification of Ilex kudingcha samples.Neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,isochlorogenic acid B,isochlorogenic acid A,isochlorogenic acid C,caffeic acid and isoquercetin have good antioxidant activity.Cluster analysis showed that Ilex kudingcha samples from different producing areas showed certain regional clustering characteristics.Themethod of content determination is good.CONCLUSION By integrating the fingerprint and antioxidant activity of Ilex kudingcha,the antioxidant activity contribution of each component in Ilex kudingcha was determined,to provide experimental basis for elucidating the antioxidant activity substance base and quality control method of in Ilex kudingcha.
