摘 要:优化胶体磨复合酶法浸提香菇中香菇多糖的工艺。以香菇多糖提取率为评价指标,采用胶体磨复合酶法,经单因素试验、正交试验和工艺验证,优化其关键工艺参数并比较。结果显示,优化后的参数为料液比1︰40 (g/m L)、酶添加量2.5%、酶解温度50℃、酶解时间40 min。在此条件下香菇多糖提取率为21.59%,提取效率比其他提取方法提高1.3~3.3倍(P<0.05)。该工艺节能环保、条件温和,为香菇多糖的产业化优化提供参考。To optimize the extraction process of lentinan from Lentinus edodes by colloid mill combined with enzyme,with the extraction rate of lentinan as the evaluation index,the key process parameters of lentinan were optimized and compared through single factor test,orthogonal test and process validation by using colloid mill compound enzyme method.The results showed that the optimized parameters were as follows:the liquid ratio 1:40(g/mL),the amount of enzyme added 2.5%,the temperature of enzymatic hydrolysis 50℃,and the time of enzymatic hydrolysis 40 min.Under this condition,the extraction rate of Lentinus edodes polysaccharide was 21.59%,and the extraction efficiency was 1.3-3.3 times higher than that of other extraction methods(P<0.05).The process is energy saving,environmental protection and mild conditions,which will provide a new reference for the optimization of lentinan industrialization.
