摘 要:为提高大麦苗麦绿素的提取效率,充分利用资源,在水提法的基础上改良使用微波和添加纤维素酶辅助提取麦绿素。结合单因素试验,采用响应面法优化麦绿素提取工艺,进一步测定麦绿素中生物活性组分含量,并对麦绿素的抗氧化活性进行分析。结果表明,大麦苗麦绿素的最佳工艺条件为料水比1∶8.7,纤维素酶添加量0.7%,微波处理时间6.9 min。在此工艺条件下进行三次麦绿素提取试验,得到麦绿素提取率的平均值为23.14%,与预测值接近。麦绿素中总多酚与总黄酮、可溶性多糖及蛋白、叶绿素、SOD酶活、甜菜碱含量分别为34.74 mg GAE/g、47.44 mg RE/g、249.81 mg/g、179.66 mg/g、24.63 mg/g、418.82 U/g、35.33 mg/g。麦绿素对水溶性ABTS+·、脂溶性DPPH·和·OH清除率的IC_(50)值分别为19.93、57.58、2605.32 mg/L。综上,证明了响应面法优化改良麦绿素提取工艺的可行性,并对麦绿素的提取及成分探究提供一定的参考依据。In order to improve the extraction efficiency of barley green pigment from barley seedlings and make full use of resources,microwave-assisted extraction and cellulase assisted extraction of barley green pigment were used on the basis of water extraction.Combined with single factor experiment,the extraction process of barley green pigment was optimized by response surface methodology.The content of bioactive components in barley green was further determined,and the antioxidant activity of barley green was analyzed.The results showed that the optimum technological conditions of barley green were as follows:the ratio of material to water was 1∶8.7,the addition of cellulase was 0.7%,and the microwave treatment time was 6.9 min.The average extraction rate of barley green was 23.14%,which was close to the predicted value.The contents of total polyphenols,total flavonoids,soluble polysaccharides,protein,chlorophyll,SOD activity and betaine in barley green were 34.74 mg GAE/g,47.44 mg RE/g,249.81 mg/g,179.66 mg/g,24.63 mg/g,418.82 U/g and 35.33 mg/g,respectively.The IC_(50) values of the clearance rates of water-soluble ABTS+·,liposoluble DPPH·and·OH by wheat green were 19.93,57.58 and 2605.32 mg/L,respectively.In conclusion,the feasibility of optimizing and improving the extraction process of barley green pigment by response surface methodology was proved.A certain reference basis is provided for the extraction and composition exploration of barley green pigment.
