摘  要:番茄红素是一种天然功能性类胡萝卜素,因化学稳定性及水溶性较差降低了生物利用率,将其制备成靶向脂质体可改善其水溶性并扩大其应用范围。实验采用探头式超声-逆向蒸发法制备以硬脂醇半乳糖苷为配体修饰的番茄红素肝靶向脂质体,单因素及响应面法优化后最佳工艺为大豆卵磷脂与胆固醇比7∶1、配体与固体脂质比1∶40、药脂比1∶48、旋蒸时间35 min、旋蒸温度51℃,在此条件下制备的脂质体包封率为(91.13±0.20)%,载药量为(2.39±0.37)%,粒径为(252.63±5.87)nm,在不同光照条件、贮藏温度以及有无氧气条件下稳定性均良好。动物实验显示番茄红素肝靶向脂质体组小鼠肝脏部位番茄红素含量最高,证明了经硬脂醇半乳糖苷修饰后,番茄红素脂质体肝靶向性增强。Lycopene is a natural functional carotenoid,but its bioavailability is reduced due to poor chemical stability and water solubility,so it is prepared into targeted liposomes to improve its water solubility and expand its application scope.Probe ultrasound-reverse evaporation method was used to prepare lycopene liver-targeting liposomes modified with stearic alcohol galactoside as ligand.The process was optimized by single factor test and response surface methodology as follows:soybean lecithin/cholesterol ratio 7∶1,ligand/solid lipid ratio 1∶40,drug/lipid ratio1∶48,rotation time 35 min,and rotation temperature 51℃.Under these conditions,the encapsulation efficiency of the prepared liposomes was(91.13±0.20)%,the drug loading was(2.39±0.37)%,and the particle size was(252.63±5.87)nm.The stability was good under different light conditions,storage temperatures and oxygen conditions.Animal experiments showed that the lycopene content in the liver of mice in the lycopene liver targeting liposomes group was the highest,which proved that the liver targeting of lycopene liposomes was enhanced after stearyl alcohol galactoside modification.
