摘 要:采用超声辅助低共熔溶剂法对滇黄精黄酮进行提取工艺研究,并初步研究其生物学活性。在单因素氯化胆碱-乳酸摩尔比、超声提取温度、超声提取时间、氯化胆碱-乳酸含水量和料液比对滇黄精黄酮提取率的试验基础上,选取超声提取温度、超声提取时间和氯化胆碱-乳酸含水量进行响应面优化试验以研究其最佳提取工艺。为研究其生物活性,选择抗氧化和降血糖试验对其进行评价。结果表明滇黄精黄酮最佳工艺条件为:氯化胆碱-乳酸摩尔比1∶2、超声提取温度45℃、超声提取时间40 min、氯化胆碱-乳酸含水量20%和料液比1∶20 g/mL,在此条件下滇黄精黄酮提取率为17.13%±0.25%。滇黄精黄酮具有较强的清除DPPH自由基和ABTS自由基能力、抑制α-葡萄糖苷酶和α-淀粉酶活性,其EC50值分别为2.11、1.63、1.70、2.69 mg/mL,表明滇黄精黄酮具有一定的抗氧化和降血糖能力。滇黄精黄酮具有较强的生物学活性,可以作为一种食品行业可用的原料。The extraction technology of flavonoid from polygonatum kingianum was studied by ultrasonic-assisted deep eutectic solvents method,and its biological activity was preliminarily studied.On the basis of single factor experiments of choline chloride-lactic acid molar ratio,ultrasonic extraction temperature,ultrasonic extraction time,choline chloride-lactic acid water content and material liquid ratio,the response surface optimization experiment was conducted to select ultrasonic extraction temperature,ultrasonic extraction time and choline chloride-lactic acid water content for optimal extraction process.Antioxidant and hypoglycemic tests were selected to evaluate biological activities.The results showed that the optimum extraction conditions of flavonoid from Polygonatum kingianum was as follows:choline chloride-lactic acid molar ratio of 1∶2,ultrasonic extraction temperature of 45℃,ultrasonic extraction time of 40 min,choline chloride-lactic acid water content of 20%and material liquid ratio of 1∶20 g/mL.Under these conditions,the extraction rate of flavonoid from Polygonatum kingianum was 17.13%±0.25%.Flavonoid from Polygonatum kingianum had strong ability to scavenge DPPH free radicals and ABTS free radicals and to inhibitα-glucosidase andα-amylase activity.The EC50 values of its activity were 2.11,1.63,1.70 and 2.69 mg/mL,respectively.The results showed that flavonoid from Polygonatum kingianum had certain antioxidation and hypoglycemic effects.Flavonoid from Polygonatum kingianum had strong bioactivity and could be used as a raw material in the food industry.
