摘 要:采用超声辅助提取玉米须多糖(corn silk polysaccharides,CSP),并经H2O_(2)-VC体系降解制备3种低分子质量多糖:CSP-1、CSP-2、CSP-3,同时研究不同分子量多糖降血脂活性及其机制。结果表明,CSP、CSP-1、CSP-2和CSP-3的抑制胰脂肪酶活性和与胆酸盐(甘氨胆酸钠和牛磺胆酸钠)结合作用在一定浓度内线性相关,其EC_(50)值分别为3.72、7.54和6.46 mg/mL,3.27、5.92和5.23 mg/mL,5.63、2.75和4.90mg/mL,3.51、3.01和2.18 mg/mL;EC_(50)值按大小排序为CSP> CSP-1> CSP-2> CSP-3,表明分子量越小其抑制胰脂肪酶活性和越好,即CSP-3具有较强的降血脂能力。采用高血脂小鼠模型研究CSP-3体内降血脂机制,结果表明其降血脂的机制主要是调节血脂和抗氧化平衡,血脂平衡是通过提高LPL和HL酶活性以降低TC、TG和LDL-C含量及提高HDL-C含量来实现,抗氧化系统平衡是提高SOD、GSH-px和CAT抗氧化酶类活性来实现的。低分子量玉米须多糖CSP-3是一种潜在的具有很强降血脂活性的功能性食品添加剂。Polysaccharides(CSP)was extracted from corn silk by ultrasonic-assisted hot water extraction,and degraded by combination of ascorbic acid and H2O2 into three low-molecular-mass polysaccharides(CSP-1,CSP-2 and CSP-3).Its hypolipidemic and mechanisms were studied.The results showed that the inhibition of pancreatic lipase activity by CSP,CSP-1,CSP-2 and CSP-3 and their binding with bile salts(sodium glycylcholic acid and sodium taurocholate)were linearly correlated within a certain concentration.Their EC_(50) values were 3.72,7.54 and 6.46 mg/mL,3.27,5.92 and 5.23 mg/mL,5.63,2.75 and 4.90 mg/mL,3.51,3.01 and 2.18 mg/mL,respectively.The order of magnitude of EC_(50) values were:CSP>CSP-1>CSP-2>CSP-3,indicating that smaller molecular weight CSP-3 had better hypolipidemic ability.The hyperlipidemic mice model was used to study the mechanism of CSP-3 in lowering blood lipid.The results showed that mechanism of hypolipidemic was to regulate the balance of blood lipid and antioxidation by increasing activities of LPL and HL enzymes to reduce the contents of TC,TG and LDL-C and increasing the contents of HDL-C.The balance of antioxidation system was achieved by increasing the activities of SOD,GSH px and CAT antioxidant enzymes.Low molecular weight corn silk polysaccharide CSP-3 can be a potential functional food additive with strong hypoglycemic.
