摘  要:在单因素试验基础上,利用响应面法优化聚酰胺和大孔吸附树脂纯化竹叶花椒黄酮的工艺,考察酰胺-大孔吸附树脂用量比、上样质量浓度、上样液流速、洗脱剂浓度、洗脱剂流速、洗脱剂体积对竹叶花椒黄酮纯化效果的影响,同时通过DPPH自由基和ABTS自由基的清除试验对竹叶花椒黄酮的抗氧化活性进行评估。结果表明,聚酰胺-大孔吸附树脂用量比3︰5(g/g)、上样质量浓度6 mg/mL、上样液流速1.5 mL/min、洗脱剂60%乙醇溶液、洗脱剂流速2 mL/min、洗脱剂体积4 BV,在此条件下得到总黄酮平均含量为87.35%。竹叶花椒黄酮对DPPH自由基和ABTS自由基均具有一定清除作用,试验结果可为进一步研究竹叶花椒药用价值提供参考。On the basic of single factor experiment,the response surface methodology was used to optimize the purification process of flavonoids from Zanthoxylum armatum DC with polyamide and macroporous adsorption resin.The effects of the ratio of amide to macroporous adsorption resin,the concentration of loading sample,the flow rate of loading sample,the concentration of eluent,the flow rate of eluant,and the volume of eluant on the purification of flavonoids from Zanthoxylum armatum DC were investigated.At the same time,the antioxidant activity of Zanthoxylum armatum DC flavonoids was evaluated by DPPH radical and ABTS radical scavenging tests.The results showed that the ratio of polyamide to macroporous adsorption resin was 3︰5(g/g),the concentration of loading solution was 6 mg/mL,the flow rate of loading solution was 1.5 mL/min,the concentration of eluent was 60%ethanol solution,the flow rate of eluant was 2 mL/min,and the volume of eluent was 4 BV.Under this condition,the average content of total flavonoids was 87.35%.The flavonoids of Zanthoxylum armatum DC have certain scavenging effects on DPPH free radicals and ABTS free radicals,to provide a reference for further research on the medicinal value of Zanthoxylum armatum DC.
