基于甘蔗糖蜜的黑曲霉产β-Ffase酶的单因素条件研究

摘  要:为了探明在甘蔗糖蜜体系下黑曲霉产β-Ffase酶的变化规律,本研究开展单因素实验。实验结果表明,在糖蜜添加量15%、氮源酵母粉20g/L、pH5.5、孢子接种量10%、温度30℃的黑曲霉菌株产β-Ffase发酵条件下,培养48h后,β-Ffase的酶活为463.87U/mL,结合表面活性剂吐温-80,与相关文献提供的培养条件相比,提高1.6倍,细胞干重为11.77 g/L,提高1.48倍。In order to investigate the variation of Aspergillus niger enzyme production in the sugarcane molasses system,we used single factor optimization experiment in this study.The results showed that the optimum fermentation conditions forβ-Ffase production of Aspergillus niger strains were as follows:15%molasses,20 g/L nitrogenous yeast powder,pH 5.5,10%spore inoculation,and temperature 30℃.After 48 h culture,β-Ffase enzyme activity was 463.87 U/mL,combined with surfactant Tween-80,1.6 times higher than the culture conditions provided by the literature,and the cell dry weight was 11.77 g/L,1.48 times higher.
