摘 要:在植物活性成分的提取中,超声辅助法是近年来兴起的一种绿色技术。以乙醇为提取液,采用超声波辅助提取法提取山楂黄酮。通过BBD响应面优化设计,进一步研究了山楂黄酮的提取效率,同时用DPPH法分析了该工艺下山楂黄酮提取物的结构活性,以抑制率表示。实验结果显示,实验条件:41.24∶1(mL/g)的液料比、69.99%的乙醇体积分数和38.93 min的萃取时间为该实验最佳工艺条件,此条件下黄酮提取率达到1.123 mg/g。DPPH法的实验结果表明,其半抑制浓度IC50=0.147 mg/mL,具有良好的抗氧化性能,说明超声辅助法在山楂黄酮类化合物的提取过程中能较大程度地保留黄酮类化合物的结构活性。In the extraction of plant active ingredients,ultrasound-assisted method is a green technology that has emerged in recent years.In this experiment,hawthorn flavonoids were extracted by ultrasonic-assisted extraction method using ethanol as the extraction solution.The extraction efficiency of hawthorn flavonoids was further investigated by BBD response surface optimization design.The structural activity of hawthorn flavonoid extracts under this process was also analyzed by DPPH method,expressed as inhibition rate.The results showed that the experimental conditions:41.24∶1(mL/g)liquid-to-material ratio,69.99%ethanol volume fraction and 38.93 min extraction time were found to be the best process conditions for this experiment,which could achieve a relatively good flavonoid extraction rate of 1.123 mg/g.The experimental results of the DPPH method showed that its semi-inhibitory concentration IC50=0.147 mg/mL,with good antioxidant properties,indicating that the ultrasound-assisted method can retain the structural activity of flavonoids to a greater extent during the extraction of hawthorn flavonoids.
