摘 要:含有sbe3-rs基因型的高抗性淀粉含量水稻品种(系)具有调节餐后血糖、改善血脂和增强饱腹感等功效。以‘降糖稻1号’为sbe3-rs基因供体亲本通过杂交育种和回交转育方法培育优质高产高抗性淀粉水稻新品种具有重要意义。sbe3-rs基因第16个外显子的第105位处有T→C的碱基突变,基于该突变位点开发高通量KASP分子标记,利用该分子标记对杂交F_(1)后代50个单株和回交BC_(1)F_(1)后代44个单株共94份材料进行基因分型检测,结果显示50份F1样品中有47个单株基因型为C/T杂合基因型,44份BC_(1)F_(1)群体材料中共有28个单株基因型为C/T杂合基因型。利用已开发的成熟的sbe3-rs的CAPS分子标记检测验证结果完全一致。同时利用开发的KASP分子标记对杂交F_(2)分离群体进行基因分型鉴定,对应高抗性淀粉基因型单株成熟期收获进行抗性淀粉含量测定,结果完全符合。结果表明针对sbe3-rs的突变位点开发KASP分子标记应用杂交回交群体,克服了前期开发的CAPS分子标记步骤繁琐缺点,同时具有高通量、低成本和高准确性等优点,可以高效应用于分子标记辅助选育高抗性淀粉水稻品种改良育种工作当中。A series of rice varieties containing the sbe3-rs gene with high resistant starch content,such as‘Jiangtangdao I’,have the ability to regulate postprandial blood glucose,improve blood lipids and enhance satiety.It is important to develop new rice varieties with high quality,high yield and high resistant starch by cross breeding and backcrossing using'Jiangtangdao I'as the donor parent of sbe3-rs gene.The KASP molecular marker was used to genotype 50 individual plants of the F,progeny and 44 individual plants of the BC_(1)F_(1)progeny.The results showed that 47 plants out of 50 F_(1)samples were of C/T heterozygous genotype,and a total of 28 plants out of 44 BC_(1)F_(1)population materials were of C/T heterozygous genotype.The CAPS molecular marker assay using the well-developed sbe3-rs was validated and the results were in complete agreement.At the same time,the developed KASP molecular marker was used for genotyping of F_(2) segregation population,and the RS content of each plant corresponding to the high resistant starch genotype was measured at maturity stage,and the results were all in complete agreement.The results showed that the development of KASP molecular markers for the sbe3-rs mutation locus can be applied to the hybrid backcross population,which overcomes the disadvantages of the previously developed CAPS molecular markers and has the advantages of high throughput,low cost and high accuracy,and can be efficiently applied to the breeding work of molecular marker-assisted selection for the improvement of highly resistant starchrice varieties.
