摘  要:该研究采用稀释平板法从“酸汤子”自然发酵玉米液中分离筛选乳酸菌,采用分子生物学技术对其进行菌种鉴定,并对其生长特性及耐酸耐胆盐性能、α-淀粉酶抑制活性、抗氧化活性和抑菌性能等益生特性进行研究。结果表明,共分离得到2株乳酸菌菌株,编号为YM-1、YM-2,经鉴定分别为植物乳杆菌(Lactiplantibacillus plantarum)和乳酸片球菌(Pediococcus acidilactici)。菌株YM-1、YM-2均具有稳定的生长状态与良好的产酸能力;在pH 2.0和0.3%胆盐条件下培养均可存活;无细胞上清液(CFS)的α-淀粉酶抑制率分别为72.57%与79.52%;CFS、细胞悬液(IC)和无细胞提取物(CFE)均有较强的1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除能力,CFS和CFE具有总抗氧化能力和铁离子还原能力;对金黄色葡萄球菌(Staphylococcus aureus)、志贺氏菌(Shigella)、大肠杆菌(Escherichia coli)、沙门氏菌(Salmonella)均具有抑制作用,其中菌株YM-2对金黄色葡萄球菌的抑制能力最大,抑菌圈直径达到20.77 mm。In this study,lactic acid bacteria were isolated and screened from'Suan tangzi'naturally fermented corn liquid by dilution plate method,identified by molecular biology technique,and the growth characteristics and probiotic properties(acid and bile salt resistance,α-amylase inhibitory activity,antioxidant activity and antimicrobial activity)were studied.The results showed that 2 strains of lactic acid bacteria were isolated,numbered as YM-1 and YM-2,and identified as Lactiplantibacillus plantarum and Pediococcus acidilactici.The strains YM-1 and YM-2 had stable growth situation and excellent acid production ability,could survive under the conditions of pH 2.0 and 0.3%bile salt.The inhibition rates ofα-amylase in cell-free supernatants(CFS)were 72.57%and 79.52%,respectively.The CFS,intact cells(IC)and cell-free extracts(CFE)of both strains had strong 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical scavenging ability,CFS and CFE had total antioxidant capacity and iron ion reducing capacity.The two strains had inhibitory effects on Staphylococcus aureus,Shigella,Escherichia coli and Salmonella,among them,the strain YM-2 had stronger inhibitory ability,and the diameter of inhibition zone reached 20.77 mm.
