摘 要:本研究以长叶竹柏叶片为材料,对其进行叶绿体基因组测序及构建系统发育树,分析长叶竹柏叶绿体基因组的结构特征。结果表明,长叶竹柏的叶绿体基因组结构(GenBank No.OL435123)缺乏反向重复(IR)区,叶绿体基因组大小为133870 bp,总GC含量为37%,共注释基因数为119个,包括80个蛋白编码基因,35个tRNA基因和4个rRNA基因。叶绿体基因组注释显示,大多数的基因与光合作用及基因转录翻译相关,其中43个光合作用相关基因、64个自我复制基因、8个其他蛋白编码基因和4个未知功能基因。密码子偏好性分析表明,共检测出44623个密码子,亮氨酸(Leu)是长叶竹柏叶绿体基组中占比最大的密码子,占10.54%,密码子偏向使用A/U两种碱基。长叶竹柏叶绿体基因组有49个SSRs,其中包含26个单核苷酸、22个二核苷酸和1个三核苷酸3种类型的SSRs。系统进化树显示,长叶竹柏(Nageia fleuryi,OL435123.1)与竹柏(Nageia nagi,AB830885.1和LC572146.1)聚在同一支,具有较近的亲缘关系。In this study,the leaves of Nageia fleuryi used as materials,the chloroplast genome of Nageia fleuryi was sequenced and a phylogenetic tree was constructed to analyze the structural characteristics.The results showed that the chloroplast genome structure of Nageia fleuryi(GenBank No.OL435123)lacked the reverse repeat(IR)region.The chloroplast genome was 133870 bp in length,the total GC content was 37%,and the total number of innotated genes was 119,including 80 protein-coding genes,35 tRNA genes and 4 rRNA genes.The chloroplast genome annotation showed that most of the genes were related to photosynthesis,gene transcription and translation,including 43 photosynthesis related genes,64 self-replicating genes,8 other coding protein genes and 4 unknown functional genes.The codon preference analysis showed that a total of 44623 codons were detected,and leucine(Leu)was the largest codon in the chloroplast base group of Nageia fleuryi,accounting for 10.54%,and the codon prefered to use A/U bases.There were 49 SSRs in the chloroplast genome of Nageia fleuryi,contained 26 single nucleotide,22 dinucleotide and 1 trinucleotide SSRs.The phylogenetic analysis indicated that the Nageia fleuryi(OL435123.1)and the Nageia nagi(AB830885.1 and LC572146.1)were clustered in the same branch and had a relatively close genetic relationship.
