摘 要:为了开发低成本高品质的天然黑色素,采用平板初筛和摇瓶复筛从实验室保存的38株产黑色素卡胞链霉菌中筛选到高产菌株HM3。通过单因素试验和响应面优化确定最佳发酵工艺为葡萄糖1%、蛋白胨0.33%、酵母浸粉0.67%、K_(2)HPO_(4)·3H_(2)O 1.07%、酪氨酸0.79%、CuSO_(4)0.00003 mol/L、pH 7.5,温度28℃、接种量3%,摇瓶装液量100 m L/500 mL。优化后黑色素产量达到5.8 g/L,较优化前提高了4.27倍。HM3黑色素的紫外、红外吸收光谱具有与真黑素一致的特征。HM3黑色素具有较好的光、热、氧化剂和还原剂稳定性;对DPPH自由基清除率为81.75%,ABTS自由基清除率为78.42%,超氧自由基清除率为57.98%,羟基自由基清除率为36.51%,具有较好的体外抗氧化活性。研究结果为HM3黑色素的发酵生产和进一步应用提供理论依据。To develop nature melanin with high-quality and low-cost,the high-yield Streptomyces capoamus strain HM3 was screened from 38 strains preserved in the laboratory by plate and shaking flask fermentation.Using single factor experiments and response surface method,the fermentation conditions of HM3 were determined as follows:glucose 1%,peptone 0.33%,yeast extract 0.67%,K_(2)HPO_(4)·3H_(2)O 1.07%,Tyr 0.79%,CuSO_(4)0.00003mol/L,pH 7.5,temperature 28℃,inoculum size 3%,medium volume 100 mL/500mL.Under the above conditions,melanin yield by HM3 fermentation reached 5.8 g/L,which was increased by 4.27 times than before optimization.The ultraviolet and infrared absorption spectra of HM3 melanin are consistent with that of eumelanin.HM3 melanin had good stability under light,heat,oxidant and reducing agent.Its DPPH radical scavenging rate was 81.75%,ABTS+radical scavenging rate was 78.42%,superoxide radical scavenging rate was 57.98% and hydroxyl radical scavenging rate was 36.51%.All the above showed the good antioxidant activity in vitro.The results provide a theoretical basis for melanin production by HM3 fermentation and further application of melanin.
