摘 要:本文采用高效液相色谱法测定饮料中叶黄素酯的含量。样品经氯化钠和无水乙醇分散,石油醚萃取出叶黄素酯,在氢氧化钾-异丙醇溶液中60℃水浴皂化成游离的叶黄素,再经正己烷萃取,无水乙醇稀释后使用YMC类胡萝卜素色谱柱分离,使用紫外检测器于445 nm测定吸光度,外标法定量。结果表明,在0.0515~5.1500 mg·L^(-1)线性关系良好,相关系数大于0.9999;方法检出限为5.35 mg·kg^(-1),方法定量限为17.80 mg·kg^(-1);按照优化后的样品处理方法同一水平加标,加标回收率为93.7%~101.0%;进行重复性实验,精密度为2.24%。该方法具有良好的回收率、重现性和精密度,针对性强,适用于饮料中叶黄素酯含量的检测,可为叶黄素酯的测定研究和市场监管提供参考。In this paper,the content of lutein ester in beverage was determined by high performance liquid chromatography.The sample was dispersed with sodium chloride and anhydrous ethanol,the lutein ester was extracted from petroleum ether,and then saponified into free lutein in potassium hydroxide-isopropyl alcohol solution in a water bath at 60℃,then extracted by n-hexane,diluted by anhydrous ethanol,and separated by YMC carotenoid column.Absorbance was measured at 445 nm by UV detector and quantified by external standard method.The results showed that the linear relationship between 0.0515 and 5.1500 mg·L^(-1)is good,and the correlation coefficient is greater than 0.9999.The limit of detection was 5.35 mg·L^(-1),and the limit of quantitation was 17.80 mg·L^(-1).According to the optimized sample processing method,the same level of spiking was used,and the spiking recovery rate was 93.7%~101.0%;Conduct repetitive experiments with a precision of 2.24%.The method has good recovery rate,repeatability,precision and pertinence.It is suitable for the determination of lutein ester in drinks,which can provides reference for the determination of lutein ester and market regulation.
