摘 要:该研究旨在优化超声辅助低共熔溶剂提取新疆大果沙枣多酚类化合物的工艺,评价其体外抗氧化活性。首先比较12种不同组分的低共熔溶剂和3种传统溶剂,从中筛选出得率最高的低共熔溶剂。然后通过单因素试验和响应面Box-Behnken设计试验优化超声辅助低共熔溶剂提取大果沙枣多酚和黄酮的同时考察DPPH自由基、ABTS阳离子自由基清除能力。结果显示,氯化胆碱-苹果酸摩尔比为1∶1、含水率25%、料液比1∶48(g∶mL)、超声时间34 min、超声温度57℃时,大果沙枣多酚得率63.378 mg/g,黄酮得率为33.528 mg/g,DPPH自由基清除能力为35.004 mg TE/g,ABTS阳离子自由基清除能力为82.443 mg TE/g。通过不同提取方法对大果沙枣多酚、黄酮得率及抗氧化活性的比较,发现它们之间呈正相关关系(P<0.05)。该研究提供了一种绿色、高效的从大果沙枣中提取多酚类化合物的方法,在提高了活性成分得率的同时保证了抗氧化活性,适用于大果沙枣活性成分和抗氧化活性的同时提取,对新疆南疆地区大果沙枣资源开发具有科学意义。The technology of ultrasound-assisted extraction of polyphenols components from Elaeagnus moorcroftii Wall.ex Schlecht.in Xinjiang by deep eutectic solvent was optimized and its antioxidant activity in vitro was evaluated.Firstly,12 different kinds of eutectic solvents and 3 traditional solvents were compared to screen out the low eutectic solvent with the highest yield.Then,a single factor test and response surface Box-Behnken design test were used to optimize the extraction of polyphenols and flavonoids by ultrasounds-assisted deep eutectic solvent and to investigate the free radical scavenging ability of DPPH free radicals and ABTS cationic radicals.Results showed that when the molar ratio of choline chloride to malic acid was 1∶1,the moisture content was 25%,the ratio of material to liquid was 1∶48(g∶mL),the ultrasonic time was 34 min,and the ultrasonic temperature was 57℃,the yield of polyphenols was 63.378 mg/g and the yield of flavonoids was 33.528 mg.Through the comparison of different extraction methods on the yield of polyphenols and flavonoids and antioxidant activity,it was found that there was a positive correlation between them(P<0.05).This study provides a green and efficient method for extracting antioxidant components from Elaeagnus moorcroftii Wall.ex Schlecht.,which not only improves the yield of active components but also ensures the antioxidant activity,and is suitable for the extraction of active components and antioxidant activity in the Elaeagnus moorcroftii Wall.ex Schlecht.at the same time.It has scientific significance for the development of Elaeagnus moorcroftii Wall.ex Schlecht.resources in southern Xinjiang.
