摘 要:目的:基于分光光度法的反应机理,建立一种便携、低毒、保质期长的亚硝酸盐快速检测方法。方法:采用3种有机酸分别代替国标法中的浓盐酸作为酸性介质,并优化3组试剂[柠檬酸组(CA组)、酒石酸组(TA组)、2-羟基丁二酸组(MA组)]中有机酸的最佳用量,进行对比研究。同时,在3组试剂中加入茶多酚作为抗氧化剂,考察不同浓度的茶多酚对3组试剂存储效果的影响。结果:3组试剂中柠檬酸、酒石酸、2-羟基丁二酸均可替代国标法中的浓盐酸作为酸性介质,时效性强;CA组、TA组、MA组线性范围分别为0.05~20.00 mg·L^(-1)、0.2~10.0 mg·L^(-1)、0.2~10.0 mg·L^(-1),28℃时存储时间分别为180 d以上、60 d、60 d。3组试剂下,食品中亚硝酸盐都有较高的加标回收率,相对标准偏差均小于5%;使用国标法和该方法对同一样品进行检测,结果一致性良好。结论:3组试剂中的有机酸均可代替浓盐酸作为酸性介质,安全性和时效性均有提高,加入茶多酚后3组试剂的存储稳定性均有提高,其中CA组的存储时间可达180 d,在亚硝酸盐实际检测工作中更加实用。Objective:To establish a portable,low toxicity,and long shelf life rapid detection method for nitrite based on the reaction mechanism of spectrophotometry.Method:Three types of organic acids were used to replace the concentrated hydrochloric acid in the national standard method as the acidic medium,and the optimal dosage of organic acids in three groups of reagents[citric acid group(CA group),tartaric acid group(TA group),and 2-hydroxysuccinic acid group(MA group)]was optimized and compared.At the same time,tea polyphenols were added as antioxidants to three sets of reagents to investigate the effect of different concentrations of tea polyphenols on the storage efficiency of the three sets of reagents.Result:Citric acid,tartaric acid,and 2-hydroxysuccinic acid in the three groups of reagents can all replace the concentrated hydrochloric acid in the national standard method as the acidic medium,with strong timeliness;The linear ranges of CA group,TA group,and MA group were 0.05~20.00 mg·L^(-1)、0.2~10.0 mg·L^(-1)、0.2~10.0 mg·L^(-1).The storage time at 28℃was above 180 days,60 days,and 60 days,respectively.Under three sets of reagents,nitrite in food has a high recovery rate with relative standard deviation less than 5%;the national standard method and this method were used to detect the same sample,and the results showed good consistency.Conclusion:Organic acids in all three groups of reagents can replace concentrated hydrochloric acid as an acidic medium,with improved safety and timeliness.After the addition of tea polyphenols,the storage stability of all three groups of reagents is improved,with the storage time of CA group reaching 180 days,making it more practical in the actual detection of nitrite.
